马尾藻多糖的提取及免疫研究

【摘要】 [目的]研究马尾藻多糖的提取工艺和药理免疫能力。[方法]以广西北海涠洲岛特有的马尾藻为材料,探索马尾藻多糖的最佳提取工艺,并对其进行免疫试验。[结果]结果表明,马尾藻多糖提取的最佳条件为:水浴温度80℃,提取时间10 h,胶体磨处理马尾藻2 min,藻粉∶水=1∶20,提取液的浓缩液体积数∶95%酒精=1.0∶3.0。马尾藻多糖对小鼠免疫的试验表明,多糖剂量为200、400、600 mg/kg.d时,小鼠脾的重量显著增加(P<0.05);当多糖剂量为400、600 mg/kg.d时,小鼠腋下淋巴结的重量显著增加(P<0.05),对单核巨噬细胞的吞噬功能有显著影响,能明显提高小鼠碳粒廓清速率,提高吞噬指数α(P<0.05),并对巨噬细胞增生以及淋巴小结形成有明显作用;当多糖剂量为600 mg/kg.d时,小鼠淋巴结不但产生明显的淋巴小结,而且形成大量巨噬细胞。[结论]该研究结果为马尾藻生理活性物质多糖的更进一步研究提供参考。

【Abstract】 [Objective]The research aimed to study the extraction process of polysaccharides from sargassum and its immunological experiment.[Method] With the spectific sargassum species in Weizhoudao,Beihai in Guangxi as experimental materials,the optimum technical conditions of extracting polysaccharides from sargassum were explored,and the immunological experiment of sargassum polysaccharides was tested.[Result]The results indicated that the optimum technical conditions of extracting polysaccharides from sargassum as follows: water-bath temperature 80 ℃,extraction time 10 h,the time of treating sargassum with colloid grinding was 2 min,the suitable proportion of algae powder and water was 1∶20 and the ration of the concentrated liquid volume number and 95% alcohol was 1.0∶3.0.Immunological experiments showed that sargassum polysaccharides could increase the weight of the spleen of mice significantly(P<0.05) when the dose of polysaccharides reached 200,400 and 600 mg/kg·d.It could significantly increased the weight of the lymphonodi axillares,strengthened the phagocytosis function of monocytle-macrophages,raised the clearance rate of mice charcoal particles,and promote the index of phagocytic(a) significantly(P<0.05),and had significant effects on the macrophages hyperplasia and the shape of lymph nodule when the dose of polysaccharides was 400,600 mg/kg·d.The significant lymph nodule in lymph node was founded,but also a large number of macrophages were formed when the dose of polysaccharides was 600 mg/kg·d.[Conclusion] This research will provide the referenced basis for the further study of sargassum physiological activity materials-polysaccharides.
