EGF·Insulin和IGF-I对食蟹猴耳部成纤维细胞体外增殖的影响
Effects of EGF,Insulin and IGF-I on the Proliferation of Fibroblast Cells in the Ear of Macaca fascicularis in vitro
【摘要】 探讨了培养基中不同浓度血清情况下,分别添加不同浓度的表皮生长因子(EGF)、胰岛素(Insulin)和胰岛素样生长因子-I(IGF-I)对食蟹猴耳部成纤维细胞体外增殖的影响,目的是寻找这3种物质促进体外培养成纤维细胞的最佳浓度,改善细胞体外培养系统,为以后体细胞及干细胞培养提供参考。将处于对数生长期的10~15代细胞进行常规消化混匀,计数,接种,培养24 h后分别向孔中加入含5%和10%的两种血清浓度、添加不同浓度的EGFI、nsulin和IGF-I的培养基,6 d后分别向孔中加入MTT,然后用酶联免疫检测仪测定各孔在492 nm处的吸光度值(OD)。血清浓度为10%情况下,培养基中添加10 ng/ml EGF、10μg/ml Isulin1、00 ng/ml IGF-I时,食蟹猴耳部成纤维细胞增殖能力最好。
【Abstract】 The objective of this study was to discuss the effects of supplementation of different concentration of EGF,insulin and IGF-I for cell proliferation of Macaca fascicularis ear fibroblast cells under 5% and 10% FBS concentrations of serum by using colorimetric MTT assay.The results showed that: when the serum concentration was 5% in DMEM,the addition of EGF(20-80 ng/ml),insulin(15 μg/ml) and IGF-I(50-100 ng/ml) had significant promoting effects on the growth of the cells respectively(P<0.05);when the serum concentration was 10% in DMEM,the addition of EGF(10-40 ng/ml),insulin(10-15 μg/ml) and IGF-I(25-100 ng/ml)had significant promotion effects on the growth of the cells respectively(gPg<0.05).
