栒子组织培养快繁技术研究
Tissue Culture and Rapid Propagation of Cotoneaster bullatus
【摘要】 [目的]为了建立栒子的组织培养快繁体系。[方法]取栒子的侧芽消毒后,接种到1/2 MS培养基上,10 d后将生长健壮的栒子侧芽分别接种到含有不同浓度激素的MS培养基上,培养30 d后,从中筛选出增殖率最高的培养基配方;后将增殖出的芽切下,接种到含有不同浓度激素的伸长培养基上进行伸长培养,30 d后,从中筛选效果最好的培养基配方;将伸长后的芽接切下,接种到含有不同浓度生长激素的1/2 MS培养基上,培养60 d后,从中筛选生根效果最好的培养基。[结果]栒子增殖率最高的培养基配方为MS+6-BA 0.5 mg/L+IAA 0.5 mg/L;伸长培养效果最好的培养基配方为MS+6-BA 0.5 mg/L+IAA 1.0 mg/L;生根效果最好的培养基为1/2 MS+NAA 0.5 mg/L。[结论]该研究可以为栒子的繁殖推广奠定基础。
【Abstract】 [Objective]The research aimed to establish the system of Cotoneaster bullatus tissue culture and rapid propagation.[Method]The shoots of C.bullatus were disinfected and inoculated on 1/2 MS medium and the buds were used for tissue culture after 10 d.The shoots of C.bullatus were regenerated from the disinfected buds on MS medium containing hormones of different concentration.And the optimal media were selected.The regenerated buds were inoculated on MS medium containing hormones of different concentration for elongation.After 30 d,the optimal media were selected.The buds of elongated were cut down and were inoculated on MS medium containing hormones of different concentration for rooting.After 60 d,the optimal media were selected.[Result]The experimental results suggested that the optimal media with highest proliferation rate were MS+6-BA 0.5 mg/L+IAA 0.5 mg/L.The optimal media with best elongation effects were MS+6-BA 0.5 mg/L+IAA 1.0 mg/L.The optimal media with best rooting effects were 1/2 MS+NAA 0.5 mg/L.[Conclusion]This research could provide basis for the propagation and extension of C.bullatus.
