紫花地丁中黄酮化合物提取方法的研究
Study on Extraction Method of Flavonoids Compound from Viola philippcia Cav
【摘要】 [目的]优化紫花地丁中黄酮化合物的提取条件,为紫花地丁的开发利用提供理论依据。[方法]以紫花地丁为试验材料,在碱提取法、乙醇提取法和微波提取法比较的基础上,通过正交试验选择最佳提取法。在最佳提取条件下,测定了紫花地丁中根、茎、花中黄酮化合物的含量。[结果]3种提取方法中,微波提取法提取时间最短,提取率效果最好,为3.65%。正交试验得到的最佳提取工艺为微波1 300 W提取20 s、乙醇浓度65%、料液比1∶15。在最佳提取条件下,全草黄酮化合物提取率最高,为4.84%;紫花地丁中根、茎、花中黄酮化合物的含量分别为4.81%、4.84%、4.90%。[结论]微波提取法是从紫花地丁中提取黄酮的最佳方法。
【Abstract】 [Objective] The objective was to optimize the extraction condition of flavonoids compound from Viola philippcia Cav and provide the theoretical basis for the development and utilization of V.philippcia.[Method] With V.philippcia as experimental material,on the basis of comparison on alkali extraction method,enthanol extraction method and microwave extraction method,the optimum extraction method was selected by orthogonal test.Under the optimum condition,the flavonoids compound content in the root,stem,flower of V.philippcia was determined.[Result] During 3 extraction methods,the extraction time of microwave extraction method was shortest and its extraction effect was the best,being 3.65%. The optimum extraction process got by orthogonal test was as follows: microwave of 1 300 W was used to extract for 20 s,ethanol concn.was 65% and the solidliquid ratio was 1∶15.Under the optimum condition,the extraction rate of flavonoids compound in whole grass was the highest,being 4.84%;the flavonoids compound content in the root,stem,flower of V.philippcia was 4.81%,4.84% and 4.90% resp.[Conclusion] The microwave extraction method was the best method for extracting flavonoids compound from V.philippcia.
