利用卡那霉素筛选非洲菊转基因植株的技术研究
Study on Selecting of Transgenic Gerbera Plants with kanamycin
【摘要】 为了能够更高效地得到非洲菊转基因植株,将非洲菊的植株接种于含有不同浓度卡那霉素的分化培养基中,当卡那霉素浓度达到25 mg/L时,外植体的生长分化完全受抑制,苗白化。将非洲菊通过愈伤组织诱导,农杆菌介导得到转基因植株,把共培养3 d的外植体进行3种不同处理。结果表明,处理3较好,即先在培养基中直接加羧苄霉素不加卡那霉素,待苗长势较佳时,再用卡那霉素筛选,PCR检测,得到7棵阳性植株,再生率为43.75%。既保证了芽诱导苗的成活率,又节省了大量提取DNA的时间和贵重药品的使用量。
【Abstract】 In order to gain more efficient Gerbera transgenic plants,Gerbera plants were inoculated with different concentrations of Km in the proliferation medium,The concention of Kanamycin at 25mg/L completely inhibited the explant growth and not shoot differentiation,young plant whitening.Through the Gerbera callus induction,Agrobacterium-mediated method to be transgenic plants,the co-cultured materials a total of 3-day explants for 3 to deal with in co-culture will be disseminated stage of the explant for 3 to deal with.The results show that a better disposal3,that is,the first increase in the medium directly Carbenicillin-carboplatin did not Kanamycin,seedling growth to be better at using kanamycin selection,pcr testing,getting 7 transgenic plants.43.75% regeneration rate.Ensured the bud induction rate,saving more time and material medica.
