转Bt+CpTI基因棉对烟粉虱种群动态影响及其几丁质酶和β-1,3-葡聚糖酶活性变化
Effect of Transgenic Bt+CpTI Cotton on Population Dynamics of Bemisia tabaci Gennadius and Activity Changes of Chitinase and β-1,3-glucanase
【摘要】 本研究调查了转Bt+CpTI基因棉SGK321棉田、其亲本常规棉SY321棉田以及间作棉田烟粉虱成虫种群的数量,测定了不同时期SGK321棉与SY321棉叶片中几丁质酶、β-1,3-葡聚糖酶的活性及蛋白质的含量。结果表明:在整个生长期间,SGK321棉田烟粉虱的种群数量一直高于SY321棉田和间作棉田。烟粉虱取食的SGK321植株叶片几丁质酶活性呈下降-上升-下降-上升的趋势;SY321上部叶片中呈下降-上升-下降-上升的趋势,其下部叶片为先下降后上升趋势;SGK321叶片中β-1,3-葡聚糖酶活性变化呈下降-上升-下降-上升趋势,而SY321叶片基本上均呈先下降后上升趋势;SY321与SGK321上部叶片中蛋白质相对含量均呈先上升后逐渐下降趋势,下部叶片与上部叶片相似但略有不同。SGK321与SY321处理植株叶片几丁质酶活性之间没有明显差异;SGK321处理植株叶片β-1,3-葡聚糖酶的活性多数时期高于SY321。探讨了棉花防御反应与棉田烟粉虱种群数量差异的相关性,以利于揭示双价抗虫棉对烟粉虱的影响及其生理防御机制,为烟粉虱的综合治理以及棉花育种等提供理论依据。
【Abstract】 The population dynamics of Bemisia tabaci Gennadius adults in SGK321,SY321 and mixed cropping fields were investigated.The activities of chitinase and β-1,3-glucanase and protein content in leaves of SGK321 and SY321 damaged by B.tabaci were determined in different time.The results showed that the population density in the SGK321 field was higher than that in the SY321 field and mixed cropping field.The activitiy trend of chitinase in SGK321 leaves was dropping-rising-dropping-rising.While that of upper and bottom leaves of SY321 were dropping-rising-dropping-rising and dropping-rising respectively.The change trends of β-1,3-glucanase activity in leaves of SGK321 and SY321 were dropping-rising-dropping-rising respectively.In upper leaves of SGK321 and SY321,the contents of protein rose firstly then dropped,which showed the similar trend in the bottom leaves.There was no significant difference between chitinase activities in leaves of SGK321 and SY321 damaged by B.tabaci.However,the activitiy of β-1,3-glucanase in SGK321 damaged by B.tabaci was higher than that in SY321 in most of periods.The relationship between the defensive reaction and the differences of B.tabaci population in SGK321 and SY321 fields were discussed.The study might reveal the effects of transgenic Bt+CpTI cotton on B.tabaci and was helpful for discovering its physiological defense mechanism and could provide theoretical basis for comprehensive control to B.tabaci and cotton breeding.
