【摘要】 从新城疫病毒(Newcastle disease virus,NDV)SGM01强毒分离株的F基因克隆出两段七肽重复序列(Heptad Repeat Region,HR)HR1和HR2,通过氨基酸序列分析显示:SGM01分离株的HR1区与F48E9、La-Sota的同源性分别为95.2%、93.7%;HR2区与F48E9、LaSota的同源性分别为89.1%、85.5%。将HR1和HR2分别插入融合表达载体pGEX-6p-1,在大肠杆菌BL21(DE3)中表达,得到预期大小的目的蛋白。这为研究HR1和HR2的结构以及其在F蛋白融合过程中所起的作用奠定了基础。更多还原

【Abstract】 Two heptad repeat fragments,HR1 and HR2,were amplified from F gene of isolated Newcastle disease virus(NDV) SGM01.The results of sequence analysis showed that the homology of HR1 with F48E9 and LaSota were 95.2% and 93.7% respectively,while those of HR2 were 89.1% and 85.5% respectively.The expected size protein was obtained by cloning HR1 and HR2 into the vector pGEX-6p-1 and expressing in E.coli BL21(DE3).These results provided a theoretical basis for the study of structure and action of HR1 and HR2 during F protein fusion.更多还原
