【摘要】 目的观察复方五味子素B(Sch-C)对人胃癌细胞株SGC-7901增殖的抑制作用并探讨其可能机制。方法MTT法观察Sch-C对人胃癌细胞的增殖抑制作用,计数法观察细胞分裂指数,流式细胞术和组化法检测细胞周期和PCNA蛋白,RT-PCR法观察CyclinD1mRNA的表达。结果Sch-C(25,50,100μg/mL)作用人胃癌细胞12,24,48h能明显抑制胃癌细胞的增殖和分裂,其中以50μg/mL Sch-C作用48h的抑制作用最为明显;50μg/mL Sch-C作用胃癌细胞48h时,流式细胞仪可以检测到凋亡峰,G0/G1期细胞明显增多且能明显抑制胃癌细胞PCNA蛋白和CyclinD1 mRNA的表达。结论Sch-C可能通过下调CyclinD1 mRNA表达阻滞细胞周期,下调PCNA蛋白表达降低细胞增殖活性来抑制人胃癌细胞株SGC-7901的增殖。更多还原

【Abstract】 Objective To observe the inhibitory effect of Schisandrim B Compound (Sch-C) on human gastric cancer SGC-7901 cells in vitro and to explore its th erapeutic mechanism. Methods Methylthiazolyltetrazolium (MTT) assay was used to examine the inhibitory effect of Sch-C on gastric cancer cells. The mitosis ind ex was assessed by using an inverted microscope. Flow cytometry and immunohistoc hemical assay was used to detect the cell cycle and PCNA protein. RT-PCR-based assay was used to detect mRNA expression of cyclinD1. Results Sch-C at the con centrations of 25,50,100 μg/mL obviously inhibited the proliferation and mito sis of gastric cancer cell after treatment for 12,24 and 48 hours. Sch-C at th e concentration of 50 μg/mL had the most obvious inhibitory efficacy after trea tment for 48 hours. An apoptotic peak was found,the percentage of the cells at G0/G1 phase of was greatly increased,and mRNA expression of PCNA protein and cy clinD1 Mrna was obviously decreased when Sch-C at the concentration of 50 μg/m L cultured with the gastric cancer cells for 48 h. Conclusion This inhibitory me chanism of Sch-C on gastric cancer cells may be associated with arresting cell cycle by down-regulating Cyclin D1 mRNA expression and lessening the proliferat ion activity of gastric cancer cells by down-regulating PCNA protein expression .更多还原
