黄芩汤对湿热型溃疡性结肠炎大鼠调节性T细胞功能的影响

【摘要】 目的:观察不同剂量黄芩汤对湿热型溃疡性结肠炎大鼠调节性T细胞(Tr细胞)功能的影响,从而探讨该方治疗该病的免疫学机制。方法:采用高脂高糖辛辣饮食干预加2,4,6-三硝基苯磺酸(TNBS)灌肠法造出湿热型溃疡性结肠炎的大鼠模型;造模成功后,分别采用大中小剂量黄芩汤、白头翁汤、柳氮磺吡啶(SASP)及生理盐水对模型进行干预;用ELISA法检测大鼠血清白介素10(IL-10)和转化生长因子β1(TGF-β1),用实时PCR(Real-tim e PCR)法检测大鼠结肠组织Foxp3 mRNA含量。结果:模型组大鼠血清IL-10、TGF-β1及结肠组织Foxp3 mRNA含量均较正常对照组明显下降(P<0.01);经干预后以上指标有不同程度回升,尤以黄芩汤中剂量组最为明显,疗效优于阳性对照药白头翁汤及SASP(P<0.05,P<0.01)。结论:黄芩汤可能通过上调以上指标而调节Tr细胞的功能,从而实现自身免疫调节的。

【Abstract】 Objective: To observe the effect of different doses of huangqin decoction on the function of regulatory T cells of ulcerative colitis rats with damp-heat syndrome,so as to explore the immunology mechanism of decoction to treat the disease.Methods: To build the rat models of ulcerative colitis with damp-heat syndrome by giving the high sugar,fat and pungent food and then clysmata with 2,4,6-trinitro-benzene-sulfonic acid.After that,the model rats were treated by giving the high,middle and small dose huangqin decoction,baitongweng decotion,sulfasalazine and normal saline.The blood serum interleukin-10 and transforming growth factorβ1 were tested by ELISA,and the Foxp3 mRNA in colon cells were tested by real-time PCR.Results: The Foxp3 mRNA,IL-10 and TGF-β1 of the model rats were significantly lower than that of normal ones.After therapy,the upper indexes increased.The indexes in the middle dose huangqin decoction group were higher that the other groups including the masculine control drugs baitongweng decoction and SASP(P<0.05,P<0.01).Conclusion: It is possible that the huangqin decoction could effectively treat UC by regulating the Tr cells and autoimmunity,which maybe the immunology mechanism of the decotion to treat the disease.
