【摘要】 目的:观察滋肾解毒中药对Ⅳ型狼疮性肾炎(LN)血清干预下肾小球系膜细胞(GMC)增殖、细胞凋亡及GMC中纤维连接蛋白(FN)mRNA表达的影响,从细胞分子生物学水平探讨滋肾解毒中药治疗LN的机制。方法:①结合血清药理学方法,分空白对照组、对照组血清+15%LN血清组、低、中、高浓度含药血清+15%LN血清组5组观察,MTT法检测GMC的增殖及抑制率,荧光法和流式细胞仪技术检测GMC细胞凋亡。②RT-PCR检测GMC中纤维连接蛋白(FN)mRNA的表达。结果:①含药血清均可明显抑制GMC增殖、诱导GMC细胞凋亡,与对照组血清相比差异有非常显著性意义(P<0·01),以高浓度组最显著。②含药血清可明显抑制GMC中FNmRNA的表达,以高浓度组最显著。结论:滋肾解毒中药的含药血清可明显抑制15%Ⅳ型LN患者血清干预下的GMC增殖、诱导细胞凋亡、抑制FNmRNA的表达,这可能是滋肾解毒中药治疗LN的分子机理。更多还原

【Abstract】 Objective:To observe the effects of Chinese crude drug of Invigorating the Kidney,Heat-clearing and Detoxicating (IKHD) on GMC proliferation and apoptosis,fibronectin(FN)mRNA expression induced by type-Ⅳ Lupus nephritis(LN).To explore the mechanism of the method of IKHD on the level of cell and molecular biology.Methods:①With the method of blood serum pharmacology,5 groups were observed:blank,control serum +15% serum of type-Ⅳ LN patients,low,medium and high concentration serum which contained Chinese medicine of IKHD +15% serum of type-Ⅳ LN patients.MTT method was adopted to detect the proliferation and inhibition ratios of GMC.Apoptosis of GMC were detected with the fluorometric method and flow cytometer respectively.②Afterwards,the method of RT-PCR was used to detect expression of FNmRNA.Results:①Different concentration serum containing traditional Chinese medicine of IKHD could inhibit the proliferation of GMC and induce the apoptosis of GMC more notably than the control group,showing extremely significant difference(P<0.01).The effect of high concentration serum was the most notable.②Different concentration serum containing traditional Chinese medicine of IKHD could inhibit the expression of FNmRNA.The effect of high concentration serum was the most notable.Conclusion:Being intervened by the 15% serum of type-Ⅳ LN patients,the serum containing traditional Chinese medicine of IKHD can inhibit the proliferation of GMC,induce the apoptosis of GMC,and inhibit the expression of FNmRNA.This may be the molecular mechanism of TCM of IKHD curing LN.更多还原
