育泡饮对自身免疫性卵巢损伤小鼠卵巢颗粒细胞的超微结构影响

【摘要】 目的:观察育泡饮对自身免疫性卵巢损伤小鼠卵巢颗粒细胞的超微结构影响。方法:以成熟的SD雌性大鼠卵巢组织制备卵巢抗原,皮下多点注射免疫BALB/C雌性小鼠,建立卵巢早衰模型。在3次免疫后连续灌服育泡饮,以强的松为阳性对照,电镜观察小鼠卵巢超微结构变化。结果:模型组颗粒细胞胞体变小,各种细胞器明显减少,细胞核染色质凝聚,核膜消失,线粒体肿胀,嵴减少,紊乱,内质网扩张。育泡饮各剂量组可见颗粒细胞体积增大,胞质内细胞器数量增多,线粒体、内质网等结构大体正常,未见染色质凝聚。结论:育泡饮具有防止颗粒细胞凋亡的作用,能抑制颗粒细胞凋亡。

【Abstract】 Objective: To assess the effects of Yupao Yin on the ultrastructure of ovarian granular cells in mice with autoimmune ovarian injury.Methods: BALB/C female mice model of POF was established by multiple sites subcutaneous injection of ovarian antigen elicited from ovarian tissue of mature SD female rats. The mice were treated with YupaoYin after 3 times of injection, with prednisone as positive control. Ultrastructural changes of ovarian granular cells were observed under electron microscope. Results: The mice in the model group had smaller granular cells with less cellular organs and assembled chromatin without caryotheca. Swollen,decreasing or disappearance of inner cristae of mitochondria and distinct dilatation of endoplasmic reticulum were seen .While in the Yupao Yin treated groups granular cells became larger, and the number of cell organs in cytoplasms increased. The morphogenesis of the mitochondria, endoplasmic reticulums became normal. No assembled chromatin was found. Conclusion: Yupao Yin can protect the granular cells from apoptosis.
