脾阴虚痴呆证病结合模型建立及滋补脾阴方药干预的实验研究

【摘要】 目的:为有效模拟临床,建立脾阴虚痴呆证病结合大鼠模型,并观察滋补脾阴(Zi-Bu-Pi-Yin,ZBPY)方药对其影响。方法:采用经典方法建立脾阴虚模型,随后将凝集态β-淀粉样蛋白1-40(β-Amyloid1-40,Aβ1-40)定位注射到双侧海马,应用ZBPY方药干预,观察各组体征变化。通过跳台仪和Morris水迷宫进行行为学测试,比较各组学习记忆能力的改变。结果:脾阴虚组和脾阴虚痴呆组表现出饮水量增加(P<0.01),肛温升高(P<0.01)等阴虚内热征象;痴呆组和脾阴虚痴呆组学习记忆力降低(P<0.05);脾阴虚痴呆+ZBPY组阴虚表现和学习记忆力均有所改善(P<0.05)。结论:本研究在完善阿尔茨海默病的证病结合模型方面作了尝试,观察了证、病、证病结合模型在体征和行为学方面的变化;ZBPY方药对脾阴虚痴呆大鼠的阴虚表现及学习记忆能力均有一定改善作用。

【Abstract】 Objective:In order to correspond to disease with syndrome in clinic more effectively,it is our aim to establish a rat model with both Alzheimer’s disease(AD) and spleen deficiency syndrome,then to study the effect of Zi-Bu-Pi-Yin(ZBPY)Decoction on it.Methods:Rats model of spleen Yin deficiency was set up by the classical way.Then the incubated Aβ1-40 was injected into hippocampus of each rat.The treated group was administered with ZBPY.Observe the changes of the rats at symptom in different phases;the behavioral abnormalities were investigated by step-down test and Morris water maze test.Results:The two model groups of spleen Yin deficiency had the appearance of Yin deficiency leading to endogenous heat,for example that increment in drinking water(P<0.01),objective elevation of the body temperature(P<0.01).The ability of learning and memory of the two AD groups was reduced conspicuously(P<0.05).The symptom of Yin deficiency and abnormal behavior of the ZBPY group were improved in AD rat model(P<0.05).Conclusion:Try to perfect the methods to make the models in this experiment and then the changes of behavior and symptom were observed in syndrome,disease,and combined syndrome with disease.ZBPY improved the symptom of spleen Yin deficiency and dysfunction of learning and memory in AD rat combined with Spleen Yin deficiency.
