【摘要】 目的:观察思的明(sidimine)对吗啡依赖猴戒断症状的治疗作用。方法:按剂量递增法皮下注射(s.c.)吗啡建立90 d吗啡依赖模型,d 91~d 97灌胃给予思的明(以生药计剂量分别为2.4,4.8,9.6g.kg-1.d-1,bid),在d 91~d 102(即戒断后12 d内)每日对各组吗啡依赖猴戒断症状进行评分,并称量体重,计算体重变化率(相对于d90的体重)。结果:思的明3个剂量均能不同程度降低吗啡依赖猴戒断症状评分,其效应呈剂量相关性,中剂量从戒断d3开始起效(P<0.05),高剂量从戒断d2即可显著控制戒断症状(P<0.01)。与模型组相比,思的明高剂量在戒断d 3和d 6~d 9能控制吗啡依赖猴体重的下降(P<0.05)。结论:思的明能缓解吗啡依赖猴的戒断症状,并有一定的控制体重下降的作用。更多还原

【Abstract】 Objective: To observe the therapeutic effect of sidimine on withdrawal symptoms in morphine-dependent rhesus monkeys.Methods: Morphine-dependence was induced by administration(s.c.) of morphine at gradual increasing doses for 90 days in rhesus monkeys.From d 91 to 97,sidimine(2.4,4.8 and 9.6 g·kg-1·d-1,bid) was orally given.During d 91 to d 102(12 days after withdrawal),the withdrawal symptoms and changes in body weight were recorded.Results: The scores of withdrawal symptoms were significantly reduced by three doses of sidimine in a dose-dependent manner.Sidimine showed the effect from third day of morphine withdrawal at the dose of 4.8 g·kg-1·d-1(P<0.05),and the effect reached a maximum at the dose of 9.6 g·kg-1·d-1(P<0.01).Compared with control group,weight lose was controlled by sidimine at the dose of 9.6 g·kg-1·d-1 in d 3 and d 6~9 after morphine withdrawal(P<0.05).Conclusion: Sidimine can alleviate withdrawal symptoms and control the weight loss in morphine-dependent rhesus monkey.更多还原
