【摘要】 目的探讨大黄素对不同类型急性胰腺炎(acute pancreatitis,AP)大鼠中性粒细胞(polymorphonuclear neutrophil,PMN)凋亡的影响及其可能机制。方法制作大鼠急性水肿型胰腺炎(acute edematous pancreatitis,AEP)与急性坏死型胰腺炎(acute necrotic pancreatitis,ANP)模型,使用大黄素进行治疗。采用流式细胞仪检测,蛋白质免疫印迹法等技术,观察各型AP大鼠模型PMN凋亡率及天冬氨酸特异性半胱氨酸蛋白酶(caspase-3、caspase-8)表达水平变化情况。结果大黄素治疗组PMN凋亡率及PMN表达caspase-3、caspase-8水平较同型AP组明显增加(P均<0.05);各AP组PMN凋亡率与caspase-3、caspase-8表达变化呈显著正相关(P<0.05)。结论大黄素可能通过caspase途径来诱导AP中性细胞凋亡。更多还原

【Abstract】 Objective To investigate the effect of Chinese medicine Emodin on polymorphonuclear neutrophil apoptosis in different types of acute pancreatitis in rats and the possible mechanism.MethodRat acute edematous pancreatitis models and acute necrotic pancreatitis models were developed and treated with Emodin.Alternations of the apoptosis rate of PMN and the expression of caspase-3, caspase-8 in PMN were studied with flow cytometer and Western blotting technique.ResultFollowing the treatment with Emodin, more apoptosis rate of PMN were observed and the expression of caspase-3, caspase-8 in PMN increased significantly(P <0.05).Positive correlation is found between the expression of caspase-3, caspase-8 in PMN and the apoptosis rate of PMN(P <0.05).Conclusions PMN death pattern of apoptosis was induced by Emodin.Emodin might regulate the PMN death patterns through caspase approach in acute pancreatitis.更多还原
