软骨多糖抑制小鼠乳腺癌Ca761生长转移的实验研究
Experimental study on inhibition effect of cartilage polysaccharidein growth and metastasis of mice breast cancer Ca761 cells
【摘要】 目的研究软骨多糖对615系小鼠乳腺癌Ca761的抑瘤和抗转移的作用机制。方法分别检测经软骨多糖和生理盐水处理过的615小鼠乳腺癌肺转移模型的瘤重、肺转移发生率,ELISA试剂盒测定血清中肿瘤坏死因子(TNF-α)的含量,明胶酶谱法检测瘤组织金属基质蛋白酶(MMPs)含量的变化,进行组间比较。结果软骨多糖具有明显的抑瘤作用,抑瘤率达到44%;治疗组肺转移率明显低于对照组(P<0.05);治疗组血清中TNF-α含量明显低于对照组(P<0.05);治疗组瘤组织中MMPs含量低于对照组(P<0.05)。结论软骨多糖可抑制小鼠乳腺癌Ca761生长和转移,其机制可能与其抑制小鼠TNF-α和MMPs分泌有关,为开发新的抑癌药物提供了选择。
【Abstract】 [Objective] To study the mechanism of anti-tumor and anti-metastasis of cartilage polysaccharide on breast cancer Ca761 cells in 615 mice. [Methods] Measure the mass of tumor, the incidence of pulmonary metastasis from the 615 breast cancer mouse model which was treated by cartilage polysaccharide or physiological saline separately. Tumor necrosis factor alpha (TNF-α) content in serum was tested by ELISA kit. The matrix metalloproteinases (MMPs) content in tumor was tested by gelatin zymography. All the results were compared between groups. [Results] Cartilage polysaccharide had marked anti-tumor effect, the rate of anti-tumor was 44%. The rate of pulmonary metastasis from therapeutic group was lower than model group significantly (P <0.05). TNF-α content in serum from therapeutic group was lower than model group significantly (P <0.05). MMPs content in tumor from therapeutic group was lower than model group significantly (P <0.05). [Conclusion] Cartilage polysaccharide inhibited the growth and metastasis of breast cancer Ca761 cells in mice, the mechanism may correlated with its inhibition on secretion of TNF-α and MMPs, and provided a selection for exploitation new anticancer drugs.
