地塞米松不同给药方式对胸膜炎的干预效应及副作用研究
Interfering effect and side-effect of two kinds of gavage administration of Dexamethasone on experimental pleurisy in rats
【摘要】 目的研究地塞米松(Dex)以不同灌胃给药方式对实验性胸膜炎的干预效应及副作用,为Dex的合理应用提供实验依据。方法采用大鼠角叉菜胶诱导胸膜炎模型,致炎前以Dex5mg/kg分别灌胃给药1次与连续给药7次,检测胸腔渗出液炎症反应指标及脂质过氧化损伤指标的变化,并计算脾脏系数和胸腺系数。结果Dex5mg/kg短时给药与连续给药对实验性胸膜炎均有一定干预效果,表现为渗出液中白细胞计数下降和蛋白含量减少,MDA和NO含量降低;但连续给药组大鼠的脾脏系数、胸腺系数明显低于短时给药组,体重也明显减轻。结论地塞米松两种灌胃给药方式对大鼠胸膜炎均有一定的干预作用,但Dex连续给药易发生免疫抑制、体重减轻等副作用。
【Abstract】 Objective To study anti-pleuritic effect and side-effect of Dexamethasone(Dex) at two kinds of gavage administration,and to provide an experimental basis for reasonable usage of Dex.Method A model of pleurisy was established with an intrapleural injection of carrageenan into rats.Dex(5mg/kg) was administrated by gavage to rats before injection of carrageenan.The administrated -rats of Dex were divided into two groups respectively:one group was gavaged once a day,another group was given once a day for 7 days.The total leukocyte count,protein content,the activity of SOD,the content of MDA and NO of pleuritic exudates,and spleen coefficient and pleura coefficient of the rats were measured.Result Dex at two given method could markedly reduce the total leukocyte count,protein content,the content of MDA and NO in exudate of the pleuritic rats,but spleen-coefficient and thymus-coefficient and body weight of continuous gavage rats by Dex significantly lower than that of Dex once gavage rats.Conclusion Different gavage administration of Dex showed apparent anti-inflammatory effect on experimental pleurisy,but there were the phenomena of immunosuppressed and lose weights in the rats of continuous gavage administered by Dex.
