长春市麻疹病毒流行株M和N基因特征分析
Analysis of genetic characteristics of wild-type measles viruses in Changchun
【摘要】 目的:了解2005~2006年长春市麻疹病毒的优势基因型及其核蛋白(N)、基质蛋白(M)基因序列特征。方法:采集2005~2006年长春市暴发和散发麻疹临床病例标本,使用WHO推荐的Vero/SLAM细胞进行麻疹病毒分离。用逆转录-聚合酶链反应(RT-PCR)扩增麻疹病毒分离株的核蛋白(N)基因羧基端450个核苷酸片段和基质蛋白(M)基因的188个核苷酸片段。对扩增产物的核苷酸序列进行测定和分析。结果:共分离到11株麻疹病毒野毒株。经对分离株的核蛋白(N)碳末端450个核苷酸进行序列测定和亲缘性关系比较,N基因与H1基因型代表株相比同源性为98.0%~98.4%,和其它已知麻疹病毒的22个基因代表株相比,长春市麻疹毒株在核苷酸水平上最大变异为10.9%。M基因的144 bp中,流行株均有4~11核苷酸不同于疫苗株Changchun-47,其中只有06 JL-1毒株的2个碱基突变有意义,引起氨基酸的改变。结论:2005年和2006年流行株的核酸序列有所不同,每个年份内流行株相同,而两年流行株之间有所差别,但是属于H1基因型。说明长春市近年来麻疹的流行株仍然是H1基因型。
【Abstract】 Objective:To investigate genotype of wild-type measles viruses and characteristic of their nucleoprotein gene and matrix gene isolated in Changchun during 2005~2006.Methods:Specimens were collected in an outbreak of measles and clinically suspected sporadic measles patients for virus isolation with Vero/SLAM cell lines recommended by WHO during 2005~2006.A 450 nucleotides fragment of the COOH-terminal of the nucleoprotein(N) gene and 188 nucleotides fragment of the matrix were amplified by RT-PCR and subjected to sequence.The sequences were compared with references viruses from GeneBank and phylogenetic analysis by using DNAstar software.Results:Eleven strains of measles viruses were isolated from patients who came from seven cities and counties of Changchun.Sequence analysis of the 450 nucleotides of COOH-terminal of nucleoprotein(N) genes indicated that these 11 strains belonged to H1 genotype,and N genes homogeneity was 98.0%~98.4% as compared with other viruses of Hl genotype from GeneBank.Among the 144 bp of the amplified fragment of M gene,there were different 4~11 bp between wild-type measles virus and Changchun-47.The epidemic strain was different from the strains widespread in the Africa,Europe and USA.Conclusion:Changchun circulating measles viruses belong to clade H1.
