猪H-FABP基因遗传多态性及与肌内脂肪含量的相关分析

【摘要】 采用PCR-RFLP的方法分析了松辽黑猪、军牧1号白猪、PIC猪、长白猪、大白猪、杜洛克猪和松野杂交猪共计127头猪H-FABP基因的遗传多态性。结果表明:(1)在5′-上游区的HinfⅠ多态位点上,所有试验猪种中均发现了多态性,在第二内含子的HaeⅢ多态位点上,所有被测猪种也都存在变异,在第二内含子的MspI多态位点上,松辽黑猪、长白猪和松野杂交猪仅表现为AA型,其他猪种均表现出多态性;(2)7个猪群的基因频率和基因型频率都达到了Hardy-Weinberg平衡状态(P>0.05);(3)松辽黑猪遗传变异对肌内脂肪含量的影响:HH基因型极显著高于Hh基因型(P<0.01),而Hh基因型又极显著高于hh基因型(P<0.01),最小二乘均值为:2.760>2.598>2.335,dd和Dd基因型分别极显著高于DD基因型(P<0.01),最小二乘均值为:2.763>2.732>2.347,结果提示:可通过提高"HH-dd"基因型的频率来增加松辽黑猪肌内脂肪含量,从而改善松辽黑猪的肉质。

【Abstract】 The genetic polymorphism of H-FABP gene in 127 pigs including Songliao black pig,Junmuyihao white pig,PIC pig,Landrace pig,Large White pig,Duroc pig and Songliao-Boar crossing pig was detected by PCR-RFLP.The results were as follows:(1) All the detected pig breeds varied at the Hinf Ⅰ polymorphism locus of 5’-upstream region,All the tested pig breeds varied at the Hae Ⅲ polymorphism locus of the second intron region,Songliao black pig 、Landrace pig and Songliao-Boar crossing pig only revealed AA genotype while other pig breeds revealed polymorphism at the MspI polymorphism locus of the second intron region;(2) all gene frequency and genotype frequency of seven porcine populations achieved Hardy-Weinberg equilibrium(P>0.05);(3) genetic variance’s effect on IMF content in Songliao black pig:HH was superior to Hh extreme significantly(P<0.01),and Hh superior to hh extreme significantly(P<0.01),the least squares mean:2.760>2.598>2.335,dd and Dd super to DD respectly extreme significantly(P<0.01),the least squares mean:2.763>2.732>2.347, suggesting that Songliao black pig’s meat quality could be improved by increasing HH-dd genotype frequency in porcine group.
