鸭白细胞介素2全基因组结构分析
Full-length genome organization analysis of duck interleukin 2
【摘要】 依据鸡白细胞介素2(chIL-2)全基因组序列设计引物探针,以简易基因组抽提法获得的鸭外周血单核细胞基因组DNA为模板,用长距离聚合酶链式反应(LD-PCR)扩增出鸭白细胞介素2(duIL-2)全基因片段。经序列测定,得到的duIL-2基因组DNA序列全长为3528bp。运用生物信息学方法对duIL-2的基因、启动子结构和cDNA序列编码的成熟蛋白进行系统分析,结果表明,duIL-2基因具有典型的4个外显子和3个内含子结构,和已知禽类及哺乳类同系物基因总体结构具有明显的相似性。鸭和鸡及其他哺乳动物IL-2基因的启动子序列具有显著的保守性,都具有AP-1、NF-AT、CD28RE、OCT、TATAbox转录起始因子结合位点和预测的转录起始位点。duIL-2cDNA序列编码的成熟蛋白进化分析表明,鸭和鹅的亲缘关系最近,和哺乳动物的亲缘关系较远,显示出明显的种属差异。
【Abstract】 Using a pair of special primers designed according to chicken interleukin 2 gene(IL-2)sequence and genome DNA template obtained from duck peripheral blood monocyte by simplified DNA isolation procedure,duck IL-2 gene was amplified by long distance-polymerase chain reaction(LD-PCR),with a full-length of 3 528 bp by sequencing.The gene organization,promoter and cDNA coding mature protein of duIL-2 were analyzed by bioinformatics.The results showed duIL-2 had a significant similarity with that of other avian and mammalian IL-2 genes,with four exons and three introns.There existed remarkable conserved domain in both duck and other avian and mammalian IL-2 promoter,including the transcription factor binding sites such as those AP-1,NF-AT,CD28RE,OCT,the TATA box and the predicted transcription start site.Comparative analysis of avian and mammalian IL-2 cDNAs coding mature protein revealed that duck IL-2 had a closer relationship with goose more than that of mammalian IL-2 and with obvious species specificity.
