316L不锈钢表面抗体的固定及体外细胞相容性研究

【摘要】 表面快速内皮化是预防支架植入后血栓和再狭窄的新途径。本实验分别将合成的仿生海洋生物粘性多肽及对内皮细胞特异性结合的CD34抗体固定到316L不锈钢片及支架表面,并对其体外细胞相容性进行了初步研究。结果表明:固定的多肽及抗体促进了内皮细胞在钢片表面的粘附和增殖,提高了细胞活性,增加了细胞与不锈钢基体的结合强度。提高多肽浓度,可以促进内皮细胞的粘附,但浓度过高,反而抑制细胞的粘附;提高抗体浓度,可以增加抗体的固定及细胞的粘附。支架固定抗体后,内皮化速度加快。该方法促进了材料表面的原位内皮修复速度,因此可用于支架材料及组织工程材料等的表面改性。

【Abstract】 Rapid re-endothelialization of implanted coronary artery stent is a new way for preventing both thrombus and restenosis.In this paper,adhesive polypeptide mimics of marine organism and antibodies potentially selective targeting with CD34 antigens on endothelial cells were immobilized onto 316L stainless steel foils and stents with the expect to improve the re-endothelialization process.The results showed that the immobilized polypeptide and antibodies increased the attachments,growths,viabilities,as well as retentions of cells.The immobilized polypeptide promoted the attachment of ECs,but overdose polypeptide might restrict the attachment.With the increase of antibody concentration,the immobilized antibodies and cell attachments were improved.After immobilization of polypeptide and antibody,the re-endothelialization of 316L stainless steel stents was accelerated.The method enhanced the in-situ recovery of ECs,and thus could be used to modify the surface of stent materials and tissue engineering materials.
