局部亚低温对大鼠脑内注射凝血酶后基质金属蛋白酶-9表达的影响
Effect on Expression of Matrix Metalloproteinase-9 by Injection of Thrombin in Rats Brains with Local Mild Hypothermia
【摘要】 目的探讨局部亚低温对大鼠脑内注入凝血酶后基质金属蛋白酶-9(matrix metalloproteinase-9,MMP-9)mRNA及蛋白表达的影响。方法雄性Wistar大鼠140只,随机分为脑内注射凝血酶(T)组和脑内注射凝血酶加局部亚低温(T+H)组。每组再分为对照组以及脑出血后6h、24h、48h、72h、5d、7d共7个亚组,T+H组于基底节注射凝血酶后立即给予4h的局部亚低温治疗,各亚组分别应用RT-PCR以及Western印记对MMP-9蛋白及mRNA的表达进行测定。结果T组MMP-9mRNA表达量呈升高趋势,于脑出血后48h达顶峰,T+H组MMP-9mRNA表达低于T组(P<0.01)。T组MMP-9蛋白表达量亦随时间延长呈上升趋势,48h达顶峰,然后下降,T+H组MMP-9蛋白表达较T组下降(P<0.01)。结论局部亚低温可抑制脑内注射凝血酶后MMP-9蛋白及mRNA表达的增加。
【Abstract】 Objective To explore the effect of local mild hypothermia on mRNA and protein expression of matrix metalloproteinase-9(MMP-9)by injection of thrombin in rats brains.Methods 140 male Wistar rats were randomly divided into two groups:thrombin injection(T)group,in which thrombin were stereotaxically injected into right caudate nucleus and the local mild hypothermia(T +H)group,in which the rats were given 4 h local mild hypothermia right after the injection of thrombin.Each group was divided into 7 subgroups:control,6 h,24 h,48 h,72 h,5 d and 7 d after operation.RT-PCR and Western blot were used to evaluate expression of MMP-9 mRNA and protein.Results In T group,compared with control,thrombin injection significantly increased MMP-9 mRNA and protein expression.In T+H group,compared with T group,the elevation of MMP-9 mRNA and protein expression were attenuated at all time points by hypothermia treatment(P<0.01).Conclusions These findings suggested that local mild hypothermia could reduce the deleterious consequence of thrombin by suppressing the elevation of MMP-9 mRNA and protein expression.
