局灶性脑缺血大鼠脑内MMP-2及MMP-9表达与滋补肝肾中药的干预效应

【摘要】 目的研究滋补肝肾的中药复健片对局灶性脑缺血大鼠脑内明胶酶A(MMP-2)和明胶酶B(MMP-9)表达的影响,试图探讨其治疗缺血性脑卒中的作用机制。方法雄性Wistar大鼠49只随机分为假手术组、模型组(分设1、2、3周组)、给药组(分设1、2、3周组),后两组进行右侧近端大脑中动脉电凝术造成大鼠局灶性脑缺血模型(MCAO),假手术组不凝闭大脑中动脉,其余操作同后两组。造模成功后第6天起给药组予复健片,剂量为按体重9 g/(kg.d),余两组分别予等量NS,均灌胃给药,1次/d。分别在相应时间点取材后用酶联免疫吸附法检测MCAO大鼠脑内MMP-2、MMP-9的表达。结果模型组在给药1、2、3周后组内比较差异无统计学意义;给药组在治疗2周和3周后均较同组前期比较差异有统计学意义,提示MMP-2、MMP-9活性表达随用药时间延长逐渐增强;给药组在治疗1周后MMP-2、MMP-9活性较同期模型组差异无统计学意义,但2周及3周后表达显著增强,较同期模型组差异有统计学意义。结论复健片治疗缺血性脑卒中的作用机制可能与后期上调MMP-2、MMP-9的表达从而抑制胶质瘢痕形成,以部分解除神经功能重塑的负面因素有关。

【Abstract】 Objective The experiment was drawed up to study the expression of proteinase-2(MMP-2) and proteinase-9(MMP-9) to analyze the action mechanism of Fujian tablet to cerebral ischemic stroke.Methods Male Wistar rats,49 in all,were divided into sham operated group,model group(1 week,2 weeks,3 weeks) and medication administration group(1 week,2 weeks,3 weeks).The rats of model group and medication administration group were resulted to focal cerebral ischemic model through electric coagulation in right proximate cerebrum median artery.The rats of sham operated group were also carried out craniotomy,without occlusoring cerebrum median artery.Other operations were the same as the model group.The rats of medication administration group were lavaged with Fujian tablet after 6 days.The dosage for each kilogram body weight was nine gram Chinese native medicine everyday,while other two groups were given the isometric physiological saline once daily.The effects of Fujian tablet on expression of matrix metal MMP-2 and matrix metal MMP9 in the brain of MCAO rats were observed with enzyme linked immunosorbent assay at corresponding time.Results The MMP-2 and MMP-9 had no significant deviation among the model group of one week,two weeks and three weeks.The MMP-2 and MMP-9 of medication administration group of two weeks and three weeks had respectively significant deviation with those of the early days,which illustrated the MMP-2 and MMP-9 expressed more strengthenly along with the medication.The deviations of MMP-2 and MMP-9 of medication administration group with those of the model group of one week had no statistical significance,while the deviations of medication administration group of two weeks and three weeks with those of the model group had statistical significance.Conclusions The mechanism of Fujian tablet treating cerebral ischemic stroke is that it can refrain the form of the glial scar through the up-regulation of MMP-2 and MMP-9 to remove the negative factors of remolding nerve function.
