不同强度运动对糖尿病大鼠认知功能的影响及机理探索

【摘要】 目的观察对比不同强度运动对糖尿病大鼠认知功能的影响及其机制。方法将SD大鼠随机分组,正常对照组10只,模型组30只为链脲佐菌素(streptozotocin,STZ)造模成功的1型糖尿病模型。模型组又分为阳性对照组(未游泳),1h/d游泳组和力竭游泳组,每组10只。在3个月时进行开场实验测试活动度和探索能力,Morris水迷宫测试空间搜索和学习记忆能力,免疫组织化学方法检测脑部海马区纤维连接蛋白(fibronectin,Fn),反映脑内纤维增生及退行性变情况。结果阳性对照组大鼠学习记忆与正常对照组相比显著下降(P<0.05),1h/d游泳组明显好于阳性对照组(P<0.05),力竭游泳组效果不明显(P>0.05)。正常对照组和1h/d游泳组比阳性对照组海马区脑组织Fn阳性区域显著减少(P<0.05)。结论适度运动可以改善糖尿病模型大鼠的学习记忆和探索能力,究其机理与运动可抑制脑组织退行性变密切相关。

【Abstract】 Objective To investigate the possible effects of exercise intensity on the cognitive function in diabetic rats and explore the underlying mechanisms. Methods Forty Sprague-Dawley female rats were randomly divided into 4 groups (n=10 each): control,diabetic,diabetic and 1 h/d swimming,diabetic and exhausting exercise groups. Diabetes was induced using streptozotocin. Cognitive functions were evaluated three months later. Open-field test was used to test ability of activities and exploration behavior and Morris water maze (MWM) was used to examine the changes of the abilities of space search,learning and memory in rats,respectively. Collagen fiber proliferation in the hippocampus was analyzed by immunohistochemical staining with anti-fibronectin(Fn) antibody. Results Cognitive functions (the abilities of space search,learning and memory) were significantly decreased in diabetic rats compared with normal controls. 1 h/d swimming but not exhausting exercise significantly improved diabetes induced decline of cognitive functions. Similarly,diabetes lead to the significant increase of the Fn positive immunoreactivites in the hippocampus (P<0.05). Swimming at 1 h/d significantly attenuated diabetes induced increase of Fn expression. Conclusions Proper exercise can improve the abilities of space search,learning and memory in diabetes rats probably through reducing Fn expression.
