四种支原体大鼠宫内感染实验研究

【摘要】 目的建立肺炎支原体(Mycoplasma pneumoniae,MP)、发酵支原体(Mycoplasma fermentans,Mf)、穿通支原体(Mycoplasma penetrans,MPe)及梨支原体(Mycoplasma pirum,MPi)大鼠宫内感染动物模型。方法实验组43只孕鼠分别在妊娠的第12.5、13.5、14.5天腹腔注射0.2ml(2×108-9)四种支原体(MP、Mf、MPi、MPe),注射相同剂量无菌生理盐水(11只孕鼠)为对照组。计算两组孕鼠流产、早产、死胎率。产后幼鼠饲养一个月无菌条件下解剖,取母鼠心、肝、肺、肾、子宫角及幼鼠各脏器培养。选取培养结果阳性幼鼠脏器做电镜观察。结果四组实验组的流产、早产和死胎率比较差异具有统计学意义(P<0.05,P<0.05);孕鼠子宫角和心、肝、肺、肾组织支原体检出率比较差异具有统计学意义(P<0.05,P<0.05);幼鼠脑组织和心、肝、肺、肾组织支原体检出率比较差异具有统计学意义(P<0.05,P<0.05);电镜结果显示幼鼠的心、肺、肝及脑组织内感染有支原体,并提示炎症和损伤。结论妊娠期宫内感染MP、Mf、MPi、MPe四种支原体可导致流产、早产、死胎和母鼠、幼鼠心、肺、肝、肾和脑组织支原体感染以及器官组织细胞损伤。

【Abstract】 To establish a experimental model of intra-uterine infection with mycoplasma in rat,0.2 ml(2x108-9) suspensions of Mycoplasma pneumoniae,Mycoplasma fermentans,Mycoplasma enetrans and Mycoplasma pirum were intraperitoneally injected into 43 pregnant rats at 75% of length of gestational periods(12,5,13.5,14.5 days) and intraperitoneal injection with 0.2 ml of sterile physiologic saline into pregnant rats was taken as control(n=11).The rates of abortion,premature delivery and stillbirth of the pregnant rats in both groups of experiment were calculated.Young rats one month after delivery were killed and dissected under sterile condition and their internal organs were taken for cultivation and ultrastructural structure observation under electron microscopy.It was found that significant differences in rates of abortion,premature delivery and stillbirth existed among 4 experimental groups as well as between the experimental and control group respectively(P<0.05;P<0.05) Also,significant difference existed in the isolation rate of mycoplasma from uterine horn,heart,liver,lungs and kidney of the pregnant rats infected with these 4 species of mycoplasma and in comparison with those of saline-injection controls(P<0.05;P<0.05).Under ultrastructural observation,it was found that infection with mycoplasma could be demonstrated in heart,lungs,liver and brain of the young rats with the presence of inflammatiory reactions and damages.
