绵羊包虫病基因工程疫苗(Eg95)免疫试验

【摘要】 目的验证绵羊包虫病基因工程疫苗(Eg95)对新疆羊源棘球蚴病预防效果。方法应用新西兰农牧科学院沃勒斯维尔动物研究中心提供的绵羊包虫病基因工程疫苗(Eg95),对新生新疆细毛羊、巴音布鲁克羊、山羊羔羊进行二免、三免、四免,以间接ELISA法作血清抗体动态检测,在免疫42d、180d、365d、730d进行人工感染,于感染后180d剖解试验羊;试验羊二免后在包虫病的高发区放牧饲养180d、365d730d后剖解试验羊,检查免疫组与对照组羊的肝脏和肺脏荷囊数量,统计保护率(减囊率)。结果1.Eg95有体征反应,属于正常疫苗反应;2.免疫组与对照组血清IgG的OD值之比?2;3.二免后42d,人工感染的保护率:新疆细毛羊90.5%,山羊100%;4.二免后180d,人工感染的保护率:新疆细毛羊94.6%,山羊82.2%;自然感染的保护率:新疆细毛羊88.1%,巴音布鲁克羊83.6%。5.二免后365d,人工感染的保护率:新疆细毛羊97.4%;自然感染的保护率:新疆细毛羊83.6%,巴音布鲁克羊83.3%。6.二免后730d,人工感染的保护率:新疆细毛羊86.%;自然感染的保护率:87.1%;7.三免后180d,人工感染的保护率:新疆细毛羊99.2%。8.三免后365d,人工感染的保护率:新疆细毛羊95.6%,自然感染的保护率95.1%。9.四免后365d,人工感染的保护率:新疆细毛羊97.7%,自然感染的保护率95.5%。结论Eg95对新疆羊源棘球蚴病具有预防作用,免疫程序为新生羊羔初次2次,间隔期1个月(或4周),其后每年加强一次。

【Abstract】 To confirm the prophylactic effect of the genetically engineering vaccine Eg95 on hydatid disease of sheep from Xinjiang ,the genetically engineering vaccine Eg95, supplied by Wallaceville Animal Research Center,New Zealand,was used to immunize the Xinjiang Merino lambs,Bayinbuluke lambs and lamb kids. Then,the dynamic examination on the levels of serum antibodies was observed by means of ELISA assay and the immunized sheep and goats were challenged with eggs of Echinococcus granunosa at 42,180,365 or 730 days after the 2nd,3 rd and 4 th courses of immunization. The animals were killed 180,365 or 730 days after challenge with eggs,and the amount of cyst burden in liver and lungs was determined in immunized and control groups of sheep. Meanwhile,the protection rate(the cyst reduction rate)of vaccine were thereby calculated.The results showed that (1)the symptoms occurred after immunization belonged to the normal range.(2)the ratio of serum IgG OD value of immunized and control groups of sheep≥2.(3)the protection rates of vaccine 42 days after challenge in the 2 nd course of immunization were 90.5% for Xinjiang Merino sheep and 100% for goats respectively.(4) the protection rates of vaccine 180 days after challenge in the 2 nd course of immunization were 94.6% for Xinjiang Merino sheep and 82.2% for goats respectively,whereas those of vaccine for natural infection were 88.1% for Xinjiang Merino sheep and 83.6% for Bayinbuluke lambs.(5)the protection rate of vaccine 365 days after challenge in the 2 nd course of immunization was 97.4% for Xinjiang Merino sheep ,whereas those of vaccine for natural infection were 83.6% for Xinjiang Merino sheep and 83.3% for Bayin-buluke lambs respectively.(6)the protection rate of vaccine 730 days after challenge in the 2 nd course of immunization was 86.0% for Xinjiang Merino sheep;whereas that of vaccine for natural infection was 87.1%.(7)the protection rate of vaccine 160 days after challenge in the 3 rd course of immunization was 99.2% for Xinjiang Merino sheep,(8)the protection rate of vaccine 365 days after challenge in the 3 rd course of immunization was 95.6% for Xinjiang Merono sheep,whereas that of vaccine for natural infection was 95.1%.(9)the protection rate of vaccine 365 days after challenge in the 4 th course of immunization was 97.7% for Xinjiang Merino sheep,whereas that of vaccine for natural infection was 95.5%.From the report in the present study,it is concluded that the genetic engineering vaccine EG95 exhibits prophylactic effect on hydatid disease of sheep. The manoeuvre of immnunization consists of two courses of immunization with an interval of one month or 4 weeks for lamb kids as well as an enhancement immunization every year.
