吉西他滨诱导胰腺癌细胞PANC-1中survivin基因表达的研究
Study of the survivin expression in pancreatic carcinoma cell line PANC-1 induced by gemcitabine
【摘要】 目的探讨吉西他滨诱导胰腺癌细胞PANC-1中survivin表达与化疗耐药的关系。方法胰腺癌细胞株PANC-1用1640液培养,吉西他滨的浓度为1μg/mL及10μg/mL,运用RT-PCR和West-ernblot检测survivin的表达。分析细胞中survivin的表达水平与化疗耐药的关系。结果用1μg/mL及10μg/mL浓度的吉西他滨作用胰腺癌细胞株24h和48h,survivin mRNA水平分别上升了(1.34±0.12)倍,(2.40±0.17)倍和(3.33±0.20)倍,(4.41±0.18)倍;蛋白水平上升了(1.20±0.07)倍,(1.48±0.19)倍和(2.90±0.04)倍,(4.50±0.20)倍,差异有显著性意义(P<0.05)。结论胰腺癌细胞株的化疗耐药可能通过吉西他滨的作用上调survivin的表达而增强。
【Abstract】 Objective To study the relationship between chemoresistance and the expression of survivin,induced by gemcitabine,in pancreatic carcinoma cell line PANC-1.Methods The pancreatic carcinoma cell line PANC-1 was cultured by RPMI 1640 and gemcitabine with the concentration of 1 μg /ml and 10 μg /mL respectively,and survivin expression was examined by RT-PCR and Western blot.The relationship of the survivin level of the cell and its chemoresistance to gemcitabine was analyzed.Results Tweenty-four and 48 hours after culture with gemcitabine concentration of 1 μg /ml and 10 μg /mL respectively,survivinmRNA level was increased(1.34±0.12),(2.40±0.17),and(3.33±0.20),(4.41±0.18) folds,and the protein expression was enhanced(1.20±0.07),(1.48±0.19),and(2.90±0.04),(4.50±0.20) folds respectively.There was a significant difference between the two groups(P<0.05).Conclusions The intensity of pancreatic carcinoma cell lines chemoresistance could be enhanced by the action of gemcitabine on the up-regulated expression of survivin.
