胃癌组织中过氧化物酶增殖因子活化受体与VEGF mRNA表达的相关性和意义
Significance and Correlation of Expressions of PPARγ and VEGF mRNA in Gastric Cancer
【摘要】 目的研究过氧化物酶增殖因子活化受体γ(PPARγ)和血管内皮生长因子(VEGF)在胃癌组织中表达的相关性及其意义。方法采用RT-PCR方法检测36例胃癌手术标本中PPARγ和VEGF mRNA的表达,同时选取相应的肿瘤以远正常胃黏膜作为对照。结果PPARγ mRNA在胃癌中的表达阳性率和量均高于正常胃黏膜(χ2=8.574,P=0.002;t=46.54,P=0.000);VEGF mRNA在胃癌中的表达阳性率和量也均高于正常胃黏膜(χ2=8.229,P=0.004;t=45.32,P=0.000)。PPARγmRNA表达与VEGFmRNA表达呈正相关关系(r=0.429,P=0.009);PPARγ mRNA表达与胃癌淋巴结转移有关(P=0.002),与临床病理TNM分期有关(P=0.042);VEGF mRNA表达也与胃癌淋巴结转移有关(P=0.037),与临床病理TNM分期有关(P=0.045)。结论PPARγ和VEGF在胃癌进展和转移过程中可能发挥重要作用,联合检测PPARγ和VEGF表达对胃癌患者病情判断和预后评价有一定的临床意义。
【Abstract】 Objective To investigate the significance of expressions of peroxisome proliferator-activated receptor gamma(PPARγ) and vascular endothelial growth factor(VEGF) in gastric cancer as well as their correlation.Methods Thirty-six pairs of normal mucosa and cancer specimens were obtained from patients who had undergone gastrectomy for primary gastric cancer,and the expressions of PPARγ and VEGF mRNA were detected by the reverse transcription-ploymerase chain reaction(RT-PCR).Results The positive rate and level of PPARγ mRNA expression were higher in gastric cancer tissues than those in normal gastric mucosa(χ2=8.574,P=0.002;t=46.54,P=0.000).The positive rate and level of VEGF mRNA expression were also higher in gastric cancer tissues than those in normal gastric mucosa(χ2=8.229,P=0.004;t=45.32,P=0.000).Simultaneously,the expression of PPARγ mRNA was positively correlated with that of VEGF mRNA(r=0.429,P=0.009),while the high productions of PPARγ mRNA and VEGF mRNA in gastric cancer were both associated with the lymph node metastasis and clinical stage(P=0.002,P=0.042;P=0.037,P=0.045).Conclusion PPARγ and VEGF may play an important role in the development and metastasis of gastric cancer,and united detection for PPARγ and VEGF expressions may be of value to make diagnosis,evaluating prognosis in patients with gastric cancer at the same time.
