Diquat诱导的生长猪氧化应激持续时间及适宜的应激标识
Duration and Indicators of Oxidation Stress Induced by Diquat in Growing Pigs
【摘要】 【目的】通过考察生长猪一次性腹腔注射diquat后不同时间点的生产性能和血清氧化与抗氧化指标的变化,探明氧化应激的持续时间和血清敏感指标,建立氧化应激模型。【方法】选用DLY生长猪8头,体重(36.00±2.50)kg,随机分为2组,每组4头猪。应激组按8 mg.kg-1体重一次性腹腔注射diquat溶液,对照组腹腔注射相同剂量的灭菌生理盐水。试验期35 d,在试验第0、1、2、3、7、14、21、28、35d空腹前腔静脉采血,检测血清中抗氧化能力、抗氧化酶活力以及代谢产物。【结果】Diquat极显著降低了试验前期和全期试猪的生产性能,显著或极显著降低第7、14、21、28天血清谷胱甘肽过氧化物酶(GSH-Px)和超氧化物歧化酶(SOD)活力,抑制羟自由基(.OH)能力和抗超氧阴离子自由基()能力,显著提高血清丙二醛(MDA)含量,在第7、14、21、28天血清过氧化氢酶(CAT)活性有降低趋势,过氧化氢(H2O2)含量有增加趋势。到第35天,各指标差异均不显著。【结论】按8 mg.kg-1体重剂量一次性腹腔注射diquat溶液,可诱导生长猪氧化应激,氧化应激效应可持续28 d,血清SOD、GSH-Px、CAT活性和MDA含量可作为反映氧化应激的敏感指标。
【Abstract】 【Objective】The current study aimed to explore the time response and sensitive indicatiors of oxidation stress induced by one-time intraperitoneal injection of diquat by investigating growth performance and serum anti-oxidation and oxidation parameters in order to establish an oxidation stress model for growing pigs.【Method】Eight DLY growing pigs with BW(36.00±2.50) kg were randomly allotted to two groups with 4 replicates per group and one pig per replicate.Pigs in group 1 or 2 were intraperitoneally injected with diquat at a dose of 8 mg.kg-1 BW or saline.The whole trial period was 35 days.Serum was collected on 0,1,2,3,7,14,21,28 and 35 day for determination of superoxides and anti-oxidative enzyme activities.【Result】Compared to group 2,diquat injection reduced growth performance(P<0.05) of pigs at early and whole period of the trial,and decreased serum glutathione peroxidase(GSH-Px)(P<0.01) and superoxide dismutase(SOD)(P<0.01) activity,reduced the capacity of inhibiting production of hydroxy radical(P<0.05) and superoxide anion(P<0.01),increased malondialdehyde(MDA)(P<0.05) contents.Catalase(CAT) activity also tended to reduce but hydrogen peroxide(H2O2) tended to increase on 7,14,21 and 28 day.No obvious difference was observed for all measurements on 35 day.【Conclusion】Oxidation stress resulted from intraperitoneal injection of diquat(8 mg.kg-1) would last for up to 28 days.Serum SOD,GSH-Px,CAT activities and MDA content were sensitive indicators for oxidation stress.
