基于响应面分析法优化的乌饭树叶总黄酮提取条件

【摘要】 采用单因素实验、部分析因试验以及中心组合试验、响应面分析法对传统溶剂浸渍提取具有抗氧化、抗衰老作用的乌饭树叶总黄酮的工艺进行了深入研究。试验结果表明:乙醇浓度和提取温度是影响乌饭树叶总黄酮提取量的关键因子,优化得到的乌饭树叶总黄酮的最佳工艺条件为:提取前浸泡60min,液料比50:1,乙醇浓度87%,提取温度61℃,提取时间2h,在这种条件下一次提取率可达68.12%。响应面分析方法可以进一步精确乌饭树叶中总黄酮提取条件。

【Abstract】 Single-factor experiment, fraction factorial design experiment and response surface methodology (RSM) were applied to optimize the extraction conditions of total flavone, which has antioxidant and anti-aging functions from Vaccinium bracteatum Thunb. leaf using traditional solvent extraction. The results suggested that ethanol concentration and extraction temperature should be two statistically significant factors. The optimized conditions were as follows:soaking time(60 min),the ratio (ml:g) of solvent to material(50:1), ethanol concentration(87%) , extraction temperature (61℃)and extraction time(2 h). Under the optimized operating conditions, the once extraction rate of total flavone was 68.12%. RSM can make extraction conditions of total flavone from Vaccinium bracteatum Thunb. leaf more accurate.
