oxHDL3、oxLDL诱导THP-1单核源性巨噬细胞表达IL-1β、IL-6、HLA-DR、CD86
oxHDL3 and oxLDL induce the expression of IL-1β,IL-6,HLA-DR,and CD86 in THP-1 monocyte-derived macrophages
【摘要】 目的:动脉粥样硬化是一种慢性免疫炎症性疾病,其脆性斑块的两个主要特征是含有大量的炎症免疫介质和巨噬细胞。oxHDL和oxLDL被发现存在于粥样斑块内,探索、比较其对炎症免疫介质产生的影响有助于进一步阐明As发生发展的机制。方法:用RT-PCR和流式细胞术等方法观察oxHDL3和oxLDL对THP-1单核源性巨噬细胞表达4种动脉粥样硬化相关基因(炎症免疫反应相关)IL-1β、IL-6、CD86、HLA-DR的影响。结果:THP-1单核源性巨噬细胞经oxHDL3和oxLDL处理后,IL-1β、IL-6的mRNA水平明显上调,CD86、HLA-DR的表面表达增强,CD86+、HLA-DR+细胞的百分比显著提高。而天然HDL3和LDL不具有上述效应。结论:oxHDL3和oxLDL经诱导或增强巨噬细胞表达IL-1β、IL-6、CD86和HLA-DR,可能参与动脉粥样硬化病变的形成与发展。
【Abstract】 Objective:Atherosclerosis is an immune and inflammatory disease.Abundant inflammatory and immune mediators and macrophages are two key features of atherosclerotic vulnerable plaques.Since oxHDL and oxLDL have been detected in atherosclerotic plaques,exploration and comparison of the effect of these two factors on production of inflammatory and immune mediators are much helpful to further elucidate the mechanism of atherogenesis.Therefore,the present study is aimed to investigate the influence of oxHDL3 and oxLDL in stimulation of expression of four atherosclerosis-related genes coding for interleukin-1β(IL-1β),interleukin-6(IL-6),class Ⅱ molecule HLA-DR,and costimulatory molecules CD86,which are immune and inflammatory-associated mediators,in THP-1monocyte-derived macrophages.Methods:After treated with oxHDL3 and oxLDL for 0-48 h,IL-1β and IL-6 mRNA levels in THP-1monocyte-derived macrophages were detected by RT-PCR,while surface class Ⅱ MHC and co-stimulatory molecules expression were analyzed by flow cytometry.Results:In THP-1monocyte-derived macrophages treated with oxHDL3 and oxLDL,IL-1β and IL-6 mRNA expression were significantly elevated.oxHDL3 and oxLDL also increased surface expression of HLA-DR and CD86 in a dose-and time-dependent manner.The percentage of HLA-DR+cells and CD86+cells were augmented remarkably.Neither native HDL3 nor native LDL caused an increase of IL-1β and IL-6 mRNA as well as HLA-DRand CD86 surface expression.Conclusion:oxHDL3 and oxLDL might be involved in atherogenesis via stimulating the expression of IL-1β,IL-6,CD86,and HLA-DR in THP-1monocyte-derived macrophages.
