大鼠皮层及海马脑片体外培养影响因素的研究

【摘要】 目的探讨大鼠皮层及海马脑片体外培养的影响因素和脑片培养的最佳实验条件。方法取大鼠离体皮层及海马脑片体外培养,2,3,5-三苯基氯化四氮唑(TTC)染色检测细胞活力,观察不同的培养时间、培养液中葡萄糖浓度及培养温度对脑片活性的影响,以确定最佳培养条件。结果随着脑片孵育时间的延长,脑片活力逐渐衰减,表现为TTC染色A490nm值下降。培养人工脑脊液中低糖含量及培养温度升高均会降低脑片活性。结论选择(35±0.5)℃,含1.1mmol/L葡萄糖浓度的人工脑脊液4h以内可作为脑片最佳体外培养条件。

【Abstract】 Objective To investigate influencing factor on rats’ cortex and hippocampal brain slices culture in vitro and approach the best experimental condition of brain slice culture.Methods Rats’ cortex and hippocampal brain slices were made and cultured in vitro.2,3,5-triphenyl tetrazolium chloride(TTC)staining was used for detection of cell viability.Effects of cultural periods of time,concentration of glucose as well as cultural temperature on slice viability were observed respectively to decide best cultural condition.Results The viability of brain slices attenuated gradually with temporal extension and the A490 nm value decrease.Lower concentration of glucose in artificial cerebrospinal fluid(ACSF)or higher temperature depressed slice activity.Conclusions According to experimental requirement,ACSF contained 11 mmol/L glucose could be selected for normal brain slices culture at the temperature of(35±0.5)℃ within 4 h.
