龙芽楤木总皂苷对糖尿病大鼠早期肾病结构改变的保护作用及其机制探讨
The protective effect and mechanism of total aralosides of aralia elata on diabetic rat kidney in early stage
【摘要】 目的探讨龙芽楤木总皂苷[total aralosides of aralia elata(Miq)seem,TASAES]对实验性糖尿病(DM)大鼠肾脏功能及结构保护作用的机制。方法应用链脲佐菌素(STZ)复制DM大鼠模型,于8 w时对正常对照组、模型组、不同剂量TASAES治疗组(低、中、高剂量组)DM大鼠进行尿量、肾重体重比、24 h尿微量自蛋白(mALB)测定;RT-PCR技术检测结缔组织生长因子(CTGF)在核酸水平上的转录情况;电镜观察超微结构变化。结果与正常对照组比较,8 w时模型组、低、中、高剂量TASAES组大鼠尿量、相对肾重/体重比、24 h mALB明显增加(P<0.05,P<0.01);与模型组大鼠比较,低、中、高剂量TASAES组大鼠相对肾重/体重比,24 h mALB下降(P<0.05),而血肌酐虽无明显变化,但有上升趋势;与模型组比较,中、高剂量TASAES组CTGF在分子水平表达差异有显著性(P<0.01);电镜显示:模型组肾基底膜增厚,厚薄不均,上皮细胞内线粒体空化,低、中、高剂量TASAES组明显减轻。结论龙芽楤木能有效降低DM大鼠尿微量白蛋白含量;降低CTGF在肾脏的表达;改善肾脏超微结构,防止基底膜增厚,保护肾小球滤过屏障。
【Abstract】 Objective To discuss the protective mechanism of total aralosides of aralia elata(Miq)seem(TASAES)on renal func- tion and structure of the diabetic rats.Methods The models of diabetic rats were made by an intra-abdominal injection of streptozotecin (STZ).At the time of 8 weeks,24 h urine volume,24 h urine mieroalbumin(24 h mALB)and kidney weight/body weight ratio were detec- ted in normal control,model and low,moderate and high doses of TASAES groups.The transcription of connective tissue growth factor(CT- GF)were detected by RT-PCR at molecular level;ultrastructure were observed at the same time.Results 24 h urine volume,24 h mALB and kidney weight/body weight ratio were increased obviousely in model and three dosages groups comparing with those in control group(P<0.05,P<0.01);24 h mALB and kidney weight/body weight ratio were decreased(P<0.05)and serum creatinine had ascending trend,but had no obvious changes in low,moderate and high doses of TASAES groups.CTGF expression in moderate and high doses of TASAES groups were decreased(P<0.01)comparing with those in model group.Electron microscope showed the irregularly thickened glomerular capillary basement membrane of model group and empty mitochondria in epithelial cells.The above phenomenons in low,moderate and high doses of TASAES groups were significantly lessened.Conclusions TASAES could lower the quantity of the urine mLAB of diabetic rats effectively, decrease CTGF expression in kidney,improve renal ultrastructure and prevent the glomerular capillary basemeat membrane from thickening to protect glomerulus filtration barrier.
