电针对大鼠脑梗死后内源性神经干细胞增殖迁移的影响

【摘要】 目的:研究电针对大鼠局灶性脑缺血后室下区(SVZ)神经干细胞增殖迁移的影响。方法:采用"线栓法"建立大鼠局灶性脑缺血模型,将大鼠随机分为模型组和电针组,按7、14、21天4个时间点分3个亚组,在梗死区对侧肢体取"曲池"和"足三里"二穴作为刺激部位,用电针治疗仪做连续刺激,每次15min,每天1次。免疫荧光法检测Dil+/Brdu+/NeuN+细胞和Dil+/Brdu+/GFAP+细胞的表达。结果:脑缺血后不同时相缺血侧SVZ区均有阳性细胞表达,而电针组在不同时相的阳性细胞数量均较同时相模型组有明显增加。结论:电针能促进缺血后神经干细胞的增殖和迁移,提示这种作用可能是电针治疗脑缺血的重要作用机制之一。

【Abstract】 Objective:To investigate the effect of electroacupuncture on proliferation and migration of neural stem cells in the subventricular zone(SVZ) after rat focal cerebral ischemia. Methods: Animal model was made by ligating external carotid artery and inserting a piece of nylon thread into the internal carotid artery. Rats were randomly divided into the model group and electroacupuncture group(containing 7,14 and 21days’ three time points).Quchi acupoint and zusanli acupoint of opposite body side of infarction on rat were consecutively stimulated for 15 min , once daily. The expression of Dil+/Brdu+/NeuN+ cell and Dil+/Brdu+/GFAP+ cell were detected by cell immunofluorescence method. Results: There were positive cells in the SVZ of ischemia side after cerebral ischemia. It was also found that the number of positive cells in electroacupuncture group was significantly more than that in the model group at different time points. Conclusion: Electroacupuncture can promote the proliferation and migration of neural stem cells after cerebral ischemia which might be one of the most important mechanisms of electroacupuncture treating cerebral ischemia.
