心肺复苏后血浆组织因子和组织因子途径抑制物的变化及银杏达莫的干预研究
Effects of ginkgo leaf extract and dityrieamole injection tissue factor and tissue factor pathway inhibitor in plasma of the rats after cardiopulmonary resuscitation
【摘要】 目的探讨大鼠心肺复苏后血浆中组织因子(TF)和组织因子途径抑制物(TFPI)含量改变以及银杏达莫干预后对其影响。方法成年雄性SD大鼠120只,随机分为对照组(A组)、复苏组(B组)、银杏达莫组(8 mL/kg)(C组),再分别按气管切开后(对照组)或自主循环恢复(ROSC)后(复苏组和银杏达莫组)0.5、3、6、9和24 h分为五个亚组,每组8只。建立窒息型大鼠心肺复苏模型,分别于各时间点取血,使用ELISA法测定各组大鼠血浆中TF和TFPI的含量。结果B组心肺复苏后各时间点血浆TF较A组显著升高(P<0.01),9 h达到峰值;C组心肺复苏后各时间点血浆TF含量较A组显著升高(P<0.01),较B组分别在6、9、24 h显著下降(P<0.01)。B组各时间点TFPI含量较A组均无显著变化(P>0.05),C组24 h TFPI较A组升高(P<0.05)。结论心肺复苏后血浆TF含量显著升高,但同期TFPI并无明显变化,存在明显的凝血和抗凝作用失衡;银杏达莫注射液可降低心肺复苏后血浆TF含量,提高TFPI含量,恢复凝血和抗凝的平衡。
【Abstract】 Objective To evaluate the changes of tissue factor(TF)and tissue factor pathway inhibitor(TFPI) in plasma of the rats after cardiopulmonary resuscitation(CPR)and the effect of ginkgo leaf extract and dityrieamole injection on TF and TFPI.Methods One hundred and twenty male adult SD rats were randomly divided into 3 groups : sham-operation group(A group),CPR group(B group) andginkgo leaf extract and dityrieamole group(C group).And each group was further divided into5 subgroups respectively on the base of time(0.5,3,6,9,24 h after incision of trachea or ROSC,n=8).Asphyxiation cardiac arrest model was used and samples were taken on each time point of subgroups.The level of TF and TFPI in plasma of rats were measured by enzyme linked immunosorbent assay(ELISA).Results In comparison with A group,the level of TF in B and C groups was increased significantly(P<0.01).The change of TFPI in B and C groups was not found at each time point.TF in C group was higher than that in A group(P<0.01) and lower than that in B group(P<0.01).Conclusion The increase of TF in plasma is significant,but the change of TFPI is not.The treatment with ginkgo leaf extract and dityrieamole may inhibit level of TF.There is a conspicuous unbalance between systems of coagulation and anticoagulation after CPR,and the unbalance can be normalized by ginkgo leaf extract and dityrieamole.
