弓形虫感染对妊娠中期BALB/c小鼠胎盘细胞凋亡的影响

【摘要】 目的探讨弓形虫感染对妊娠中期BALB/c小鼠胎盘细胞凋亡的影响。方法在妊娠第8d,采用腹腔注射法建立弓形虫感染的BALB/c小鼠动物模型,于妊娠第12、14、16和18d分批处死孕鼠,采用流式细胞技术检测胎盘细胞凋亡率,分光光度法测定半胱天冬氨酸蛋白酶-3(Caspase-3)活性。结果随着妊娠天数的增加,实验组和正常对照组孕鼠胎盘细胞凋亡率及Caspase-3酶的活力单位均在升高,差异均具有统计学意义(P<0.01);相同妊娠天数的比较显示,除妊娠18d的Caspase-3酶活力单位外,其余各妊娠天数的凋亡率以及酶活力单位实验组均高于正常对照组,差异有统计学意义(P<0.05)。结论妊娠中期弓形虫感染可导致孕鼠胎盘细胞凋亡明显增加,致使胎盘组织的完整性受到破坏,影响胎盘的功能,导致不良妊娠结局的发生。

【Abstract】 Objective To investigate the placental apoptosis of BALB/c mice infected with Toxoplasma gondii during the second trimester. Methods The animal models was established by intraperitoneal injection (i.p.) at the 8th day of pregnancy, and the placentas were separated at the 12th, 14th, 16th, 18th day, respectively.The apoptosis rate and the Caspase-3 activity units were determined by flow cytometry (FCM) and spectrophotometry. Results It was showed that not only the apoptosis rate and Caspase-3 activity units increased along with the increase of the pregnant days in both the experimental group and the normal control group, which had significant differences (P<0.01), but also the apoptosis rate and Caspase-3 activity units of the experimental group were higher than that of the normal control group at the same pregnant day, which the differences had statistical significance except that of the Caspase-3 activity units at the 18th day(P<0.05). Conclusion The placental apoptosis of pregnant mice infected with Toxoplasma gondii during the second trimester of BALB/c mice increases apparently compared with the normal control group, which will influence the integrity and function of the placental, and lead to the adverse pregnancy outcomes finally.
