大肠埃希菌脂多糖脂质体的制备及免疫保护实验
Preparation and immunoprotective effects of a liposomal lipopolysaccharide from Escherichia coli
【摘要】 目的制备临床分离的耐药大肠埃希菌脂多糖(LPS)脂质体并考察其免疫保护作用。方法采用逆向蒸发法制备大肠埃希菌LPS脂质体,透射电镜观察其形态,激光散射法分析其粒径分布,鲎试验法(LAL)测定其活性;用LPS脂质体免疫小鼠,以LPS 2倍完全致死剂量攻击小鼠,观察并记录结果。结果大肠埃希菌LPS脂质体常温下呈乳白色混悬液,透射电镜下呈多层圆形或椭圆形小囊,粒径为(300.195±108.932)nm;LAL法测定E.coliLPS脂质体和E.coliob55∶b5 LPS脂质体活性,均比未经脂质体包被的LPS降低99.9%;E.coliLPS脂质体和E.coliob55∶b5 LPS脂质体免疫鼠接受E.coliLPS攻击后72 h的存活率分别为91.67%和75.00%,与对照组存活率16.67%比较,差异均有统计学意义(P=0.000 322和P=0.000 614)。结论大肠埃希菌LPS脂质体制备方法可行,制备的LPS脂质体对该菌属的LPS攻击均具有一定的免疫保护作用。
【Abstract】 Objective To prepare a liposomal lipopolysaccharide(LPS) of Escherichia coli separated from clinic and investigate its immunoprotective effects.Methods Liposomal lipopolysaccharide of E.coli was prepared by the method of reverse evaporation.The shape was observed under transmission electron microscope,using laser light scattering to analyze its particle diameter distribution,using limulus test to evaluate its activity,using two times complete lethal dose of LPS to attack the mice immunized by the liposomal lipopolysaccharide and record the results.Results The liposomal lipopolysaccharide was oyster white suspl in common temperature.It was multilayer round or oval saccule under the transmission electron microscope,and its particle diameter was well-distributed about(300.195±108.932) nm.Compared with LPS,the liposome activity of E.coli LPS and E.coli ob55∶b5 LPS was decreased 99.9%;Using E.coli LPS to attack the mouse immunized by the liposome of E.coli LPS and E.coli ob55∶b5 LPS,the 72-hour survival rate is 91.67% and 75.00%,respectively,comparing with the control group the survival was 16.67%,the differences were statistical significance(P=0.000 322 and P=0.000 614).Conclusion The preparative method of this liposomal lipopolysaccharide is feasible and the liposomal lipopolysaccharide has invariably immunoprotection to its strain’s LPS attack.
