免疫层析试纸条检测感染小鼠肉汁中抗旋毛虫抗体的研究
Immunochromatographic strip for detection of anti-Trichinella antibodies in muscle juice of experimentally infected mice
【摘要】 目的建立一种肉类旋毛虫感染的快速免疫学检测方法。方法以胶体金标SPA和旋毛虫肌幼虫ES抗原制备免疫层析试纸条,对不同剂量旋毛虫幼虫感染小鼠后不同时间的血清及肉汁抗体进行检测,并与ELISA、镜检法及消化法的检测结果进行比较。结果用100、300、500条旋毛虫幼虫感染小鼠后6周,3组小鼠的血清及肉汁抗体阳性率均达100%;试纸条法对血清与肉汁的抗体检出率差异无统计学意义(χ2=0.22,P>0.05)。试纸条法和ELISA检测100条幼虫感染小鼠肉汁的阳性率分别为91.3%和100%(χ2=2.09,P>0.05),检测300、500条幼虫感染小鼠肉汁的阳性率均为100%;试纸条法和镜检法对3组小鼠感染后5~7周的肉汁和膈肌检测的阳性率均为98.6%。消化法检查每克肌肉虫荷<33条幼虫的6份标本,试纸条5份阳性,每克肌肉中含有6条幼虫时可被试纸条法检出。感染旋毛虫小鼠肌肉4℃保存1~7 d及-20℃保存1~7个月的肉汁抗体阳性率均为100%。结论免疫层析试纸条可用于新鲜肉、冷藏肉及冷冻肉中抗旋毛虫抗体的检测。
【Abstract】 Objective To establish one rapid immunological method for detection of Trichinella in meat. Methods The immunochromatographic strip was prepared by using SPA labeled with colloidal gold and T.spiralis muscle larval excretory-secretory(ES) antigens.The anti-Trichinella antibodies in serum and muscle juice from mice experimentally infected were assayed by the strip at different interval time after infection.The results of strip were compared with those of ELISA,trichinelloscopy and digestion method. Results Anti-Trichinella antibodies in muscle juice of mice infected with 100,300 and 500 larvae was detected two weeks post-infection(wpi),with antibody positive rate of 100%.The positive rate of muscle juice was not different from those of serum sample(χ2=0.34,P>0.05).When muscle juice from mice infected with 100 larvae were assayed by strip and ELISA,the positive rate was 91.3% and 95.7%,respectively(χ2=2.09,P>0.05);However,the positive rate of muscle juice from mice infected with 300,500 larvae were 100% using the two methods.The positive rate of muscle juice assayed by strip and diaphragm examined by trichinelloscopy was 98.6% in three group of mice 5-7 wpi.Out of 6 samples containing <33 larvae per gram of muscle(lpgm) by digestion method,5 samples were positive by strip.The antibodies in muscles containing 6 lpgm could be detected by the strip.The antibody positive rate of the infected mice muscles conserved at 4 ℃ for 1-7 d and at-20 ℃ for 1-7 months was 100%. Conclusion The immunochromatographic strip could be used for detection of anti-Trichinella antibodies in muscle juice from fresh,cool and frozen meat.
