紫菀酮蛋白结合率的HPLC测定
Determination of Protein Binding Rates of Shionone by HPLC
【摘要】 建立了HPLC法测定紫菀酮在大鼠血浆、人血浆和牛血清白蛋白(BSA)中的总浓度及游离药物浓度,并分别计算蛋白结合率,以及紫菀酮与人血浆和BSA中白蛋白结合药物的表观最大能力(βp)和药物-蛋白复合物的解离常数(Kdp)。结果表明体外紫菀酮与三者的蛋白结合率均较高,在人血浆和BSA中的βp分别为0.314和0.516μmol/L,Kdp分别为1.78×10-2和8×10-4μmol/g。
【Abstract】 A HPLC method was established for the determination of shionone in rat plasma,human plasma and bovine serum albumin(BSA).The protein binding rates and correlate parameters(βp and Kdp)of shionone with human plasma and BSA proteins were determined.Its in vitro protein binding rates with the three mediums were high.βp and Kdp in human plasma and BSA were 0.314,0.516μmol/L and 1.78×10-2,8×10-4μmol/g,respectively.
