【摘要】 目的:探索香烟+酒精造成股骨头缺血性坏死动物模型的新方法,观察TNF-α、iNOS的变化。方法:分别用酒精灌胃、密室熏烟、酒精灌胃+密室熏烟的方法建立大鼠股骨头缺血性坏死的模型,对大鼠模型及施加中药影响后的大鼠模型测定血清TNF-α,股骨头局部TNF-α和iNOS免疫组化指数,并进行光镜和扫描电镜观察。结果:各观察指标说明香烟+酒精可以明显造成大鼠全身和股骨头局部免疫因子改变,诱发股骨头缺血性坏死,光镜和扫描电镜亦观察到了坏死表现。结论:香烟+酒精可成功建立股骨头缺血性坏死的动物模型。更多还原

【Abstract】 Objective:To explore a new way of establishing avascular necrosis of femoral head (ANFH) model by cigarette and alcohol and to investigate the changes of tumor necrosis factor -α(TNF-α) and inducible nitric oxide synthase (iNOS) in ANFH models. Methods:Three methods were used to establish ANFH model: intragastric administration of alcohol, fumigation with cigarette, and intragastric administration of alcohol plus fumigation with cigarette. After the treatment of Chinese herbal medicine (CHM), the level of TNF-α in serum, TNF-αand iNOS in local femoral head were detected. Meanwhile, light microscope and scanning electron microscope were used to make morphology observation. Results:All data showed that cigarettes and alcohol caused obvious cytokine changes in serum and femoral head, and induced ANFH which was proved by light microscope and scanning electron microscope. Conclusion:The ANFH models can be successfully established by cigarette and alcohol.更多还原
