穿心莲有效部位药理毒理的初步研究

【摘要】 目的:初步研究穿心莲抗血栓有效部位(APNF)急性毒性及其对血小板聚集和血栓形成的影响。方法:最大给药剂量法初步观察APNF对小鼠的急性毒性;考察APNF对兔血浆体外血小板聚集的影响以及对手术结扎法引起静脉血栓形成的影响;同时还考察其对小鼠耐缺氧能力的影响。结果:未观察到APNF在小鼠体内的急性毒性反应;对小鼠耐缺氧能力的影响与普萘洛尔和复方丹参滴丸相当;不同生产批号的APNF在体外均能有效地抑制二磷酸腺苷诱导的血小板聚集;同时其能显著降低血栓质量,并与剂量呈一定相关性。结论:初步研究显示APNF可望成为一安全、有效的抗血栓药物,值得进一步探讨。

【Abstract】 OBJECTIVE To study acute toxicity and the effect on platelet aggregation and thrombus formation of active fractions from Andrographis Paniculate Ness.METHODS APNF in the largest amount was given to the mice by stomach feeding,and behavioral indexes and toxicity degree were continuously observed;the effect of APNF on platelet aggregation induced by ADP in rabbits in vitro was studied;the model of venous thrombosis was produced to investigate the antithrombotic effect of APNF.Moreover,the ability of hypoxia tolerance was determined by the hypoxia test.RESULTS No toxic effect was observed in APNF largest dosage.The increase of maximum platelet aggregation rate induced by ADP was evidently inhibited with APNF.APNF also reduced the wet and dry thrombus weight of inferior vena cava of rats,and the effect showed a trend of concentration-dependence.The ability of hypoxia tolerance of APNF was similar with propranolol and compositie salviae dropping pill.CONCLUSION APNF has high security in mice.It is suggested that APNF has the potential of antithrombosis due to its inhibition of platelet adhesion and it is worth to make a thorough study on APNF.
