枸杞多糖对糖尿病大鼠肾脏PKC-βⅡ及IV型胶原表达的影响
Effect of lycium barbarum polysaccharides on protein kinase C-βⅡ and collage IV of kidney in type 2 diabetic rats
【摘要】 目的:研究枸杞多糖对2型糖尿病大鼠肾脏PKC-βⅡ及IV型胶原的影响。方法:制备2型糖尿病大鼠模型,将大鼠分为正常对照组,单纯糖尿病组,枸杞多糖干预组,检测糖尿病成型12周时各组大鼠肾脏肥大指数、尿微量白蛋白、肾功能、肾脏形态学指标及PKC-βⅡ、IV型胶原的表达。结果:与糖尿病组相比,枸杞多糖干预组肾肥大指数、尿微量白蛋白、肾功能、肾脏形态学指标均有改善,且PKC-βⅡ、IV型胶原的表达明显减低。PKC-βⅡ、IV型胶原与肾功能、肾脏形态学指标存在正相关。结论:枸杞多糖可以通过抑制PKC-βⅡ、IV型胶原的表达起到肾脏保护作用。
【Abstract】 OBJECTIVE To study the effect of LBP on protein kinase C-βⅡ and collage IV of kidney in type 2 diabetic rats. METHODS Type 2 diabetic rats model was established. The rats were devided into 3 groups: normal group, diabetic group without intervention, diabetic group with intervention orally by lycium barbarum polysaccharides (LBP) (250 mg·kg-1·d-1). Abserve the kidney weight/body weight, urine microalbumin serum creatinine, blood urine nitrogen, the indexes of morphology, the expression of protein kinase C-βⅡ and collage IV after 12 weeks. RESULTS In LBP intervention group, the kidney weight/body weight, urine microalbumin, serum creatinine, blood urine nitrogen, the indexes of morphology were improved in comparison with diabetic rats. And the expression of protein kinase C and collage IV were reduced in comparison with diabetic rats. the kidney weight/body weight, urine microalbumin, serum creatinine, blood urine nitrogen, the indexes of morphology positively correlated with protein kinase C and collage IV.CONCLUSION LBP can protect the kidney of diabetic rats through reducing the expression of protein kinase C and collage IV.
