顺铂处理肝癌细胞SMMC-7721的蛋白质组学分析

【摘要】 目的:用蛋白质组技术分析顺铂(cisplatin,DDP)作用于肝癌SMMC-7721细胞后细胞内蛋白质组的变化。方法:采用四氮唑蓝还原反应法(MTT)测定细胞生长抑制率。顺铂(10mg.L-1)作用肝癌SMMC-7721细胞24h,分别收集顺铂处理组与对照组细胞,提取细胞内总蛋白并采用双向电泳(2-DE)分离,Image Master2D Platinum软件分析顺铂处理前后差异表达蛋白质,切取明显差异点,基质辅助激光解吸飞行时间质谱(MALDI-TOF-MS)鉴定差异蛋白。结果:顺铂可抑制肝癌SMMC-7721细胞的生长,其抑制作用具有剂量及时间依赖性。质谱鉴定出4个已知序列的差异蛋白点。结论:顺铂处理后肝癌SMMC-7721细胞内差异表达的蛋白可能与顺铂的抗肿瘤作用机制有关。

【Abstract】 OBJECTIVE To study changes of protein expression in SMMC-7721 cells administered with cisplatin(DDP) using a comparative proteomic approach.METHODS Inhibitory effects of DDP on SMMC-7721 cell line were measured by MTT test.SMMC-7721 cells were collected after being exposed to DDP(10 mg·L-1) for 24 hours,then total proteins were extracted and separated by two dimensional electrophoresis(2-DE).Spots with dominant different expression were selected by Image Master 2D Platinum software analysis and identified with matrix assisted laser desorption /ionization time of flight mass spectrometry(MALDI-TOF-MS).RESULTS DDP induced SMMC-7721 cell death was in a time and dosage dependent manner.Four different expression protein spots were identified by MALDI-TOF-MS.CONCLUSION These altered expression proteins might be related to the apoptotic mechanism of DDP-treated SMMC-7721 cells.
