N-琥珀酰壳聚糖肿瘤细胞亲和性的实验研究

【摘要】 目的通过研究不同相对分子质量N-琥珀酰壳聚糖与肿瘤细胞的亲和性,筛选出最佳靶向制剂载体。方法用化学降解法制备不同相对分子质量的壳聚糖,以其为原料制备6种N-琥珀酰壳聚糖,用异硫氰酸荧光素标记,与成纤维细胞、肺癌、乳腺癌、肝癌等肿瘤细胞共孵育,通过荧光显微镜和流式细胞分析确定其与肿瘤细胞亲和性的强弱。结果N-琥珀酰壳聚糖样本6与肿瘤细胞亲和性最强,48h荧光强度为74.22;其他1~5样本荧光强度分别为62.61,21.87,47.11,62.63,23.89。在与正常细胞和其他肿瘤细胞的对照实验中发现,样本6与A549肺癌细胞的亲和性最强,相对荧光强度高达167.06。结论N-琥珀酰壳聚糖样本6与肺癌细胞亲和性强,可作为靶向抗肺癌药物的载体。

【Abstract】 OBJECTIVE To investigate the affinity of N-succinyl-chitosan samples to cancer cells,and select the best one as the drug targeting carrier.METHODS A series of chitosans with different molecular weight were prepared by chemical degradation, then they were used to synthesize N-succinyl-chitosans labeled with fluorescein isothiocyanate(FITC).They were incubated with different kinds of cells for affinity test,such as fibroblast(2BS),lung cancer cells(A549),pancreatic carcinoma cells(PANC-1), colon carcinoma cells(HT-29),liver cancer cells(HepG-2),breast cancer cells(SK-BR-3).Fluorescence intensity of cells was determined by flow cytometry and fluorescence microscope to assess the affinity.RESULTS The N-succinyl-chitosan sample 6 showed the strongest affinity to canc.er cells,and the average fluorence intensity was 74.22 after incubation for 48 h,the others(1~5) were 62.61,21.87,47.11,62.63 and 23.89,respectively.In addition,sample 6 showed stronger affinity to lung cancer cells (A549)than other cell lines,the average fluorence intensity was 167.06.CONCLUSION The N-succinyl-chitosan sample 6 has strong affinity to lung cancer cells,and can be used as a kind of drug targeting carrier to lung cancer.
