VEGF在类风湿关节炎病人骨小梁成骨细胞中的表达及意义
Expression and Significance of VEGF in Trabecular Bone Osteoblasts in Rheumatoid Arthritis
【摘要】 目的探讨血管内皮生长因子(VEGF)在类风湿关节炎(RA)病人成骨细胞中的表达及作用机制。方法切取RA病人和外伤对照组病人股骨粗隆部松质骨块,采用免疫组织化学染色方法检测VEGF及其受体Flt-1和Flk-1在骨小梁成骨细胞中的蛋白表达。结果RA病人骨质疏松明显,骨小梁成骨细胞数目减少。RA组和对照组骨小梁成骨细胞同时表达VEGF及其受体Flt-1和Flk-1,但RA组表达明显减弱。结论血管源性因子VEGF以自分泌作用方式调节生理和病理状态下成人成骨细胞的表达,RA病人骨质疏松与VEGF作用减弱密切相关。
【Abstract】 Objective To study the expression and effect of vascular endothelial growth factor (VEGF) in osteoblasts in rheumatoid arthritis. Methods The trabecular bones were collected from lesser trochanter in normal and rheumatoid arthritis. The protein expressions of VEGF and its receptors Flt-1 and Flk-1 in trabecular bone osteoblasts were detected by immunohistochemistry. Results Obvious osteoporosis was found in femoral intertrochanter in rheumatoid arthritis,and the number of osteoblasts decreased significantly. Immunolocalization substantiated that the proteins of VEGF and its receptors Flt-1 and Flk-1 were co-expressed in trabecular bone osteoblasts in normal and rheumatoid arthritis,but the expression in rheumatoid arthritis was significantly weakened. Conclusion Acting as an important factor of angiogenesis,VEGF might play an important autocrine function in physiological and pathological adult osteoblast differentiation. The osteoporosis in rheumatoid arthritis is closely related to the weakened role of VEGF.
