雌孕激素替代治疗雷公藤多甙致雌性大鼠生殖系统损害的实验研究

【摘要】 目的通过雌、孕激素序贯性(HPT)干预治疗,探讨雌孕激素对雷公藤多甙(TWP)所致大鼠生殖系统损害的保护作用及卵巢早衰(POF)的防治方法。方法选择30只SD大鼠随机分为正常对照组(A组)、自然恢复组(B组)及雌孕激素干预组(C组),前12周B组、C组每天均给予TWP灌胃,A组给予等量溶媒;后4周C组给予HPT治疗,A、B组给予等量溶媒。为期16周。分别观察3组大鼠动情周期,生殖器官质量及组织形态学变化,性激素水平改变。结果与A组相比:B组大多动情周期缺失、不完整或延长;子宫及卵巢质量明显下降(P<0.05),结构受损。血清雌激素(E2)水平明显降低,、卵泡刺激素(FSH)、黄体生成素(LH)水平则明显升高(P<0.05)。而C组动情周期规律;子宫、卵巢的质量无下降(P>0.05);子宫各结构未见异常,E2、FSH及LH水平无明显改变(P>0.05)。结论TWP可致雌性大鼠生殖系统的损害,HPT对卵巢功能有较好的保护作用,可促进所致损害的恢复,是防治TWP所致POF的有效方法之一。

【Abstract】 Objective To investigate the protective effects of estrogen replacement therapy combined with progestogen on the female rats with impaired genital system induced by tripterygium wilfordii polyglycosidium(TWP).Methods The 30 SD female rats were randomly divided into three groups:control group,recovery group and treatment group.The process lasted for 16 weeks.The effects of TWP and HPT were evaluated according to the changes of estrous cycle,humid weight of ovaries and uterus in female rats,the changes of histological specimen with ovaries and uterus,and the levels of E2,LH and FSH in their blood.Results Compared with the control group,TWP disrupted the estrous cycle and decreased the humid weight of ovaries and uterus.The level of E2 secreted by ovary was lower and the level of LH and FSH secreted by pituitary was higher in the recovery group than those of the control group.There was significant difference between the control group and TWP group(P <0.05).Estrogen replacement therapy combined with progestogen could restrain these pathological changes produced by TWP,and there was no significant difference between the control group and the treatment group(P > 0.05).Conclusion TWP can damage the genital system of female rats.It is an effective method to use estrogen replacement therapy combined with progestogen periodically in these rats.
