IGFBP-6在维甲酸诱导的软骨细胞凋亡中的表达

【摘要】 目的:对维甲酸诱导的软骨细胞凋亡病理过程中胰岛素样生长因子结合蛋白-6(IGFBP-6)的表达规律进行研究。方法:1×10-6mol/L的全反式维甲酸(ATRA)作用体外培养兔的软骨细胞株,观察软骨细胞中Ⅱ型胶原的合成变化,检测软骨细胞株细胞凋亡率。采用实时定量PCR和Western印迹杂交分析软骨细胞中IGFBP-6 mRNA和蛋白质的表达。结果:ATRA(1×10-6mol/L)作用软骨细胞24 h,ATRA抑制了软骨细胞中Ⅱ型胶原的合成,并使软骨细胞凋亡率增加了250%(P<0.05)。ATRA使软骨细胞中IGFBP-6 mRNA和蛋白质的表达分别增加了140%和195%(P<0.05)。结论:维甲酸可能通过负性调控靶基因IGFBP-6的基因转录和翻译,发挥对软骨细胞的促凋亡作用。

【Abstract】 Aim:To investigate gene expression of insulin-like growth factor binding protein 6(IGFBP-6) in retinoic acid-induced apoptosis of rabbit chondrocytes.Methods:Chondrocytes from rabbit were cultured with all-trans retinoic acid(ATRA) at 1×10-6 mol/L for 24 hours.The synthesis of type II collagen and the cell apoptosis were measured in ATRA-treated chondrocytes.Furthermore,the expressions of IGFBP-6 mRNA and protein in chondrocytes were detected by real-time quantitative PCR and Western blotting analysis.Results:The synthesis of type II collagen was down-regulated by ATRA of 1×10-6 mol/L.Exposure to ATRA of 1×10-6 mol/L increased the cell apoptosis by 250% in comparison to the control group(P<0.05).There were 140% and 195% increases in the expression levels of IGFBP-6 mRNA and protein in ATRA-treated chondrocytes,respectively(P<0.05).Conclusion:The retinoic acid could up-regulate the genetic transcription and translation of IGFBP-6,thereby resulting in chondrocytes apoptosis in vitro.
