甲醛对暴露工人外周血淋巴细胞的遗传损伤作用

【摘要】 目的评价甲醛对暴露工人外周血淋巴细胞染色体和DNA损伤水平。方法选择65名甲醛暴露工人和非甲醛暴露工人70名为研究对象,采用甲基纤维素法和单细胞凝胶电泳法,分析其外周血淋巴细胞微核发生率和DNA断裂损伤。结果暴露组工人外周血淋巴细胞微核发生率、DNA断裂损伤的细胞数,与对照组比较,差异有统计学意义(P<0.05),并有随工龄、暴露浓度增加而增高的趋势。但甲醛浓度为0.5 mg/m3~3.11 mg/m3时,DNA断裂损伤细胞与对照组比较,差异无统计学意义(P>0.05)。结论甲醛暴露可导致工人外周血淋巴细胞染色体和DNA断裂损伤明显增高,DNA断裂可望成为暴露人群的早期效应标志物和致癌机制之一。

【Abstract】 Objective To investigate the chromosome and DNA damage in peripheral blood lymphocytes of occupational exposure populations.Methods All 65 workers occupationally exposed to formaldehyde and 70 workers without occupational formaldehyde exposure were recruited.Micronucleuis test and single cell gel electrophoresis assay were used to evaluate the chromosomal and DNA damage of peripheral blood lymphocytes.Results Our results showed that formaldehyde exposure could enhance observably the frequency of micronucleus occurrence and the DNA strand breaks in occupational exposure populations’ peripheral hemocytes,and the differences were statistically significant compared with control group(P<0.05).The increased tendencies with the increased exposure levels and seniority were found in those indices.However,no significant difference was found in the DNA strand breaks of 0.5~3.11 mg/m3 formaldehyde.Conclusion Formaldehyde has obvious genetic toxicity on occupational exposure staffs,DNA strand breaks may be an early effect biomarker and one of the carcinogenicity mechanisms of formaldehyde.
