LC-MS法测定人血浆中西替利嗪的浓度
Determination of Cetirizin in Human Plasma by LC-MS
【摘要】 目的:建立以液-质联用法测定人血浆中西替利嗪浓度的方法。方法:取血浆样品0·5mL,经乙酸乙酯-二氯甲烷(4:1)液-液萃取后进行检测,流动相为乙腈-0·1%甲酸水溶液(55:45),流速为0·5mL·min-1,柱温为25℃。采用电喷雾电离源(ESI),正离子方式下以选择反应监测(SRM)方式进行监测,用于定量分析的离子反应分别为m/z389→201(西替利嗪)和m/z515→497(内标:替米沙坦)。结果:西替利嗪检测浓度在2·5~400μg·L-1范围内线性关系良好(r=0·9993),定量下限为2·50μg·L-1;日内、日间RSD均不>9·9%;每个样品的分析时间<2·8min。结论:本方法分析速度快,灵敏度高,适用于西替利嗪的药动学研究。
【Abstract】 OBJECTIVE:To develop an LC-MS method for the determination of plasma concentration of cetirizin.MET-HODS:Plasma sample(0.5 mL) was detected after liquid-liquid extraction with acetic ether-dichloromethane(41).The mobile phase consisted of acetonitrile-water(0.1% formic acid)(5545) at a flow rate of 0.5 mL·min-1.An agilent 1100 tandem mass spectrometer equipped with ESI source was used as detector and was operated in positive ion mode.Selected reaction monitoring(SRM) using the precursor product ion combinations of m/z 389→201 and m/z 515→497 were used to quantify cetirizin and telmisartan(internal standard),respectively.RESULTS:The linear concentration range of cetirizin was 2.5~400 μg·L-1(r=0.999 3).The low limit of quantification was 2.5 μg·L-1.The intra-day and inter-day RSD were all no more than 9.9%.The analyzing time for each sample was less than 2.8 min.CONCLUSION:The method was proved to be rapid,sensitive and suitable for the pharmacokinetic study of cetirizin.
