酶解法提取甘薯多糖技术研究

【摘要】 甘薯经过酶解、热水浸提、除蛋白、透析、乙醇沉淀,得到甘薯多糖。通过SDS-PAGE电泳方法,分离得到4种纯多糖组分。利用纤维素酶水解甘薯多糖,显著提高了可溶性多糖的产量。对不同条件下甘薯多糖酶法提取的影响因素进行了研究,结果表明,利用酶法结合热水浸提甘薯多糖,提取率达到5.55%。通过正交试验得到酶解最适作用条件为:反应时间90min、纤维素酶添加量0.04%(酶活性≧10000μg/g)、反应温度30℃。

【Abstract】 By means of enzymatic action,hot water extraction,ethanol precipitation,the sweet potato polysaccharide was obtained.Then through SDS-PAGE electrophoresis,four pure polysaccharides were obtained.The results showed that yield of soluble polysaccharides raised remarkably by adding cellulase to hydrolyze sweet potatoes.The influence factors of extraction on sweet potato polysaccharides in enzymatic methods under different conditions was studied.The results showed that the extractive rate was reach to 5.55% when extracting sweet potato polysaccharides in enzymatic method combined with hot water soaking method.The best conditions of enzymatic action were determined by orthogonal test:the reacting time 90 min,adding amount of cellulase 0.04%(enzymatic activity≧10 000 μg/g)and reacting temperature 30 ℃.
