XAGE-1b基因在唾液腺腺样囊性癌转移中的作用

【摘要】 目的:探讨肿瘤睾丸抗原家族中XAGE-1b基因在唾液腺腺样囊性癌高转移细胞ACC-M中的高表达是否与其转移相关。方法:采用RNA干扰方法抑制腺样囊性癌低、高转移细胞ACC-2、ACC-M中XAGE-1b基因的表达,检测该基因表达改变对肿瘤细胞体外迁移能力以及裸鼠体内肿瘤细胞肺转移能力的影响。实验结果采用SPSS11.5软件包进行单因素方差分析。结果:XAGE-1b基因干扰质粒转染腺样囊性癌细胞后,RT-PCR验证干扰效率显示,XAGE-1b基因表达量显著下降;Boyden小室检测肿瘤细胞体外迁移能力明显下降;在裸鼠体内检测基因干扰后的肿瘤细胞肺转移能力与ACC-M相比也显著下降(P<0.01)。结论:RNA干扰作用抑制了XAGE-1b基因的表达,体外实验和体内检测中显著影响了肿瘤细胞的迁移黏附以及肺转移能力,初步证实了XAGE-1b基因在唾液腺腺样囊性癌转移方面有重要作用。
【Abstract】 PURPOSE: To investigate whether the high expression of XAGE-1b gene,a new found cancer/testis antigen, in high metastasis cell line ACC-M is related to the metastasis of salivary adenoid cystic carcinoma. METHODS: RNAi technology was employed to suppress the expression of XAGE-1b gene in ACC-2 and ACC-M cell lines. Migration capacity of the cell lines in vitro and metastatic capacity of the tumor cells to the lung of nude mice were detected.The data was analyzed with SPSS11.5 software package for one-way ANOVA. RESULTS: RT-PCR results showed the expression of XAGE-1b was significantly decreased after ACC cell lines was transfected with XAGE-1b siRNA expression plasmid; the migration capacity of the tumor cell lines in vitro was decreased significantly;and the metastatic capacity of the transfected cell lines in vivo was also decreased,there were significant differences between the transfected cell lines and ACC-M cell lines(P<0.01). CONCLUSIONS: Interference action of siRNA suppresses the expression of XAGE-1b gene, and affects the metastatic capacity of the tumor cells in vivo and in vitro. It is suggested that XAGE-1b gene plays an important role in the metastasis of adenoid cystic carcinoma and might be a diagnostic marker or therapeutic target for salivary adenoid cystic carcinoma.
